香栓菌抑制NLRP3炎症小体活化改善过敏性鼻炎 被引量:3

摘要 目的:研究不同剂量的香栓菌Trametes suaveolens不同提取物对豚草花粉诱导的小鼠季节性过敏性鼻炎(SAR)的影响。方法:将C57BL/6J小鼠随机分为9组:空白组、模型组、氟雷他定(LT)组、香栓菌水提物(TSW)高、低剂量组、香栓菌多糖(TSD)高、低剂量组、香栓菌石油醚提取物(TSPE)高、低剂量组,采用豚草花粉构建小鼠SAR模型,并对小鼠行为学评价,HE染色观察小鼠鼻黏膜组织病理学变化,ELISA检测小鼠血清和鼻腔灌洗液(NALF)中炎症因子水平。Western blot检测小鼠鼻黏膜组织中NLRP3、Caspase-1、IL-18、IL-1β蛋白表达水平。结果:与模型组相比,TSPE可减少炎症细胞浸润,降低小鼠血清中IgE、IL-18、IL-1β和NALF中IL-6、IL-18、IL-1β水平(P<0.05),显著下调鼻黏膜组织中NLRP3、Caspase-1、IL-18、IL-1β蛋白表达水平。结论:TSPE可改善豚草花粉诱导的小鼠SAR,其机制可能通过抑制小鼠鼻黏膜上皮细胞中NLRP3炎症小体活化,从而减轻炎症损伤。 Objective:To study the effects of Trametes suaveolens fruiting body different extracts in different doses on seasonal allergic rhinitis(SAR)induced by ragweed pollen in mice.Methods:C57BL/6J mice were randomly divided into 9 groups:blank group,model group,loratadine(LT)group,Trametes suaveolens fruiting body water extract(TSW)high-dose and low-dose groups,polysaccharide(TSD)high-dose and low-dose groups,petroleum ether extract(TSPE)high-dose and low-dose groups. The mouse model of SAR was established by ragweed pollen. Behavioral evaluation was performed on mice,and the histopathological changes of nasal mucosa were observed by HE staining. The levels of inflammatory factors in serum and nasal lavage fluid(NALF)were detected by ELISA. The expression levels of NLRP3,Caspase-1,IL-18 and IL-1β in nasal mucosa tissues of mice were detected by Western blot.Results:Compared with the model group,TSPEH could reduce the infiltration of inflammatory cells,decrease the levels of IgE,IL-18,IL-1β in serum and IL-6,IL-18,IL-1β in NALF(P<0.05),and could significantly down-regulate the protein expression levels of NLRP3,Caspase-1,IL-18 and IL-1β in nasal mucosa.Conclusion:TSPE could improve symptoms of SAR in mice induced by ragweed pollen,the mechanism may inhibit the activation of NLRP3 inflammasome in mouse nasal mucosal epithelial cells,thus preventing the occurrence of inflammatory response.
