系统性红斑狼疮外周血T淋巴细胞PD-1和Tim-3表达和意义

Expression and significance of PD-1 and Tim-3 on peripheral T lymphocyte of patients with systemic lupus erythematosus

目的:检测系统性红斑狼疮(SLE)患者外周血T淋巴细胞上PD-1和Tim-3表达水平,探讨其在SLE中的作用。方法:采用流式细胞术检测41例SLE患者、33例健康对照者(HC)外周血T淋巴细胞上PD-1和Tim-3的表达情况。分析SLE患者外周血T淋巴细胞上PD-1和Tim-3的共表达与实验室检查、临床表现、治疗的关系。结果:(1)SLE患者组PD-1~+T淋巴细胞百分比、Tim-3~+T淋巴细胞百分比和T淋巴细胞PD-1表达的平均荧光强度(MFI)均显著高于HC组,差异具有统计学意义;(2)SLE患者组PD-1~+Tim-3~-、PD-1~+Tim-3~+、PD-1~-Tim-3~+T淋巴细胞百分比和PD-1~+Tim-3~-T淋巴细胞PD-1表达的MFI显著高于HC组,差异具有统计学意义;(3)anti-nucleosome、anti-Ro52阳性的SLE患者PD-1~+Tim-3~-T淋巴细胞PD-1表达MFI明显高于阴性者,差异具有统计学意义;(4)PD-1~+Tim-3~+T淋巴细胞百分比与L呈负相关;PD-1~+Tim-3~-T淋巴细胞PD-1表达MFI与WBC、RBC、HGB、HCT、L、N、C3、C4呈负相关,与PLR、RDW呈正相关;(5)低红细胞血症组、贫血组SLE患者PD-1~+Tim-3~-T淋巴细胞PD-1表达MFI明显高于阴性者,差异有统计学意义;(6)治疗后SLE患者PD-1~+Tim-3~-T淋巴细胞PD-1表达MFI明显低于治疗前,差异具有统计学意义。结论:SLE患者PD-1~+Tim-3~-T淋巴细胞百分比和PD-1表达MFI明显升高,且与自身抗体的产生、疾病的严重程度和治疗密切相关。

Objective:To investigate the expressions of PD-1 and Tim-3 on peripheral T lymphocyte from systemic lupus erythematosus(SLE)patients and its role in SLE.Methods:The expressions of PD-1 and Tim-3 on peripheral T lymphocyte from 41SLE patients and 33 healthy controls(HC)were detected by flow cytometry.The correlations of PD-1 and Tim-3 expressions with laboratory indexes,clinical features and treatment were analyzed.Results:(1)The percentage of PD-1T lymphocyte,the percentage of Tim-3T lymphocyte and the mean fluorescence intensity(MFI)of PD-1 on T lymphocyte were significantly increased in SLE patients compared to healthy control group;(2)The percentage of PD-1Tim-3,PD-1Tim-3,PD-1Tim-3T lymphocyte and the MFI of PD-1on PD-1Tim-3T lymphocyte were significantly increased in SLE patients compared to HC group;(3)The MFI of PD-1 on PD-1Tim-3T lymphocyte was significantly increased in SLE patients with positive anti-nucleosome,anti-Ro52 than in corresponding negative ones;(4)The percentage of PD-1Tim-3T lymphocyte was found to negatively correlate with L.The MFI of PD-1 on PD-1Tim-3T lymphocyte were found to negatively correlate with WBC,RBC,HGB,HCT,L,N,C3,C4,and the MFI of PD-1 on PD-1Tim-3T lymphocyte were found to positively correlate with PLR,RDW;(5)The MFI of PD-1 on PD-1Tim-3T lymphocyte was significantly increased in SLE patients with erythropenia,anemia;(6)The MFI of PD-1 on PD-1Tim-3T lymphocyte was decreased after treatment.Conclusion:The percentage of PD-1Tim-3T lymphocyte and the MFI of PD-1 on PD-1Tim-3T lymphocyte were significantly increased in SLE patients compared to HC,and the MFI of PD-1 on PD-1Tim-3T lymphocyte associated with autoantibody production,the disease severity of SLE and treatment.
