摘要
目的:探究变应性鼻炎(AR)患者外周血CD4~+T淋巴miR-326、miR-155表达差异,分析甲基化干预对细胞及动物模型miR-326、miR-155表达及Th细胞因子IFN-γ的影响。方法:RT-qPCR、Western blot等技术检测AR患者相关因子表达,甲基化酶抑制剂体外干预培养淋巴细胞及AR大鼠模型,检测miR-326、miR-155及相关基因表达。结果:AR组miR-326、miR-155表达高于正常组(P<0.05),IFN-γ表达低于正常组(P<0.05),与miR-326、miR-155呈负相关。正常组维吾尔族miR-326表达高于汉族,IL-4 mRNA表达低于汉族(P<0.05),病例组维汉各指标差异无统计学意义,甲基化酶抑制剂干预T淋巴细胞,miR-326表达升高,但在动物模型鼻黏膜中变化差异无统计学意义。结论:AR患者CD4~+T淋巴细胞中miR-326、miR-155呈高表达,与低表达的IFN-γ呈负相关,miR-326、miR-155可能通过影响Th1细胞因子IFN-γ参与AR发生发展。
Objective:To investigate expression differences of miR-326 and miR-155 in peripheral blood CD4T lymphocytes of patients with allergic rhinitis(AR),and to analyze effects of methylation intervention on expressions of miR-326 and miR-155 and Th cytokine IFN-γ in cells and animal models.Methods:RT-qPCR,Western blot and other techniques were used to detect expressions of related factors in patients with AR. Methylase inhibitors were used in vitro to intervene lymphocyte culture and rat model of AR,and expressions of miR-326,miR-155 and related genes were detected.Results:Expressions of miR-326 and miR-155 in AR group were higher than those in normal group(P<0.05),while expression of IFN-γ was lower than that in normal group(P<0.05),which was negatively correlated with miR-326 and miR-155. In normal group,miR-326 was higher in Uygur than in Han,while expression of IL-4 mRNA was lower in Uygur than in Han(P<0.05). In case group,there was no statistical difference between Uygur and Han in various indicators. Expression of miR-326 was increased after intervention of methylase inhibitors in T lymphocytes,but there was no difference in nasal mucosa of animal models.Conclusion:miR-326 and miR-155 were high expressions in CD4T lymphocytes of AR patients,which is negatively correlated with low expression of IFN-γ. miR-326 and miR-155 may participate in occurrence and development of AR by affecting Th1 cytokine IFN-γ.
