IL-17A、IL-6、IFN-γ在卵巢癌的表达情况及相关性研究

Expression and correlation of IL-17A,IL-6 and IFN-γprotein in ovarian cancer

目的探讨白细胞介素(IL)-17A、IL-6、干扰素(IFN)-γ蛋白在卵巢癌组织的表达情况及相关性。方法选取2017年5月至2020年5月如皋市人民医院就诊的50例患者为研究对象,其中30例上皮性卵巢癌患者作为卵巢癌组,20例卵巢上皮性良性肿瘤患者作为良性肿瘤组,另选取同期因子宫肌瘤行全子宫+附件切除术且卵巢正常的20例患者作为对照组。苏木素-伊红(HE)染色观察3组卵巢组织病理结构,免疫组织化学染色分析卵巢组织IL-17A、IL-6、IFN-γ蛋白的表达情况,ELISA检测血清IL-17A、IL-6、IFN-γ表达水平,并分析IL-17A、IL-6、IFN-γ三者间的相关性。结果对照组卵巢组织无明显异常,良性肿瘤组卵巢有炎性细胞浸润,卵巢癌组肿瘤细胞大小不一,形态各异。IL-17A在对照组、良性肿瘤组呈弱阳性表达,在卵巢癌组呈强阳性表达,与血清表达水平一致。IL-6在对照组呈弱阳性表达,在良性肿瘤组呈中等阳性表达,在卵巢癌组呈强阳性表达,与血清表达水平一致。IFN-γ在对照组和良性肿瘤组呈强阳性表达,在卵巢癌组呈弱阳性表达,与血清表达水平一致。IL-17A和IL-6呈强正相关关系(r=0.9918),IL-17A和IFN-γ呈强负相关关系(r=-0.9774)。结论IL-17A、IL-6和IFN-γ可能协同参与了卵巢癌的发生、发展。

Objective To investigate the expression and correlation of interleukin(IL)-17A,IL-6 and interferon(IFN)-γprotein in ovarian cancer tissue.Methods A total of 50 patients treated in the People’s Hospital of Rugao from May 2017 to May 2020 were selected as the subjects,including 30 patients with epithelial ovarian cancer as the ovarian cancer group and 20 patients with benign ovarian tumor as the benign tumor group.Another 20 patients with normal ovaries who underwent the total hysterectomy plus adnexal resection for hysteromyoma in the same period were selected as the control group.Hematoxylin-eosin(HE)staining was used to observe the pathological structure of the ovarian tissues in the three groups.Immunohistochemical analysis was used to detect the expressions of IL-17A,IL-6 and IFN-γ proteins in the tissues.The enzyme-linked immunoadsordent assay(ELISA)was used to detect the expression levels of IL-17A,IL-6 and IFN-γ in serum.The correlations among IL-17A,IL-6 and IFN-γ protein were analyzed.Results There was no obvious abnormality in the ovarian tissue of the control group.The inflammatory cell infiltration was found in the benign tumor group,and the tumor cells were of different sizes and shapes in the ovarian cancer group.IL-17A was weakly positively expressed in the benign tumor group and the control group.IL-17A was strongly positively expressed in the ovarian cancer group,which was consistent with that in serum.IL-6 was weakly positively expressed in the control group,moderately positively expressed in the benign tumor group,and strongly positively expressed in the ovarian cancer group,which was consistent with that in serum.IFN-γ was strongly positively expressed in the control group and the benign tumor group,weakly positively expressed in the ovarian cancer group,which was consistent with that in serum.There was a strong positive correlation between IL-17A and IL-6(r=0.9918),and a strong negative correlation between IL-17A and IFN-γ(r=-0.9774).Conclusion IL-17A,IL-6 and IFN-γ might synergistically involve in the occurrence and process of ovarian cancer.
