松材线虫HAP基因家族鉴定及生物信息学分析 被引量:2

摘要 为了解松材线虫体内分泌的组氨酸酸性磷酸酶(HAP)在侵染寄主过程中的作用,从生物信息学角度,通过松材线虫全基因组数据,鉴定松材线虫HAP基因家族成员,对其进行理化性质、信号肽、跨膜结构域、系统发育、染色体定位、基因结构及保守基序分析。并且通过松材线虫接种黑松不同时期和被松树代谢产物β-蒎烯胁迫后的转录组数据,对松材线虫HAP基因家族成员进行基因表达谱分析,进而对不同松树代谢产物处理下松材线虫体内BxHAP基因表达定量分析。结果表明:鉴定出的34个松材线虫HAP基因家族成员多数为酸性蛋白,12个成员为典型效应因子;34个成员分属于6个不同的亚家族,均匀分布在松材线虫前6条染色体上,基因结构和保守基序在各亚家族内部保持相对一致;BxHAP基因在松材线虫侵染松树过程中发挥重要功能,具有影响松树代谢产物的能力,其中BxHAP5在松材线虫抵抗松树代谢产物过程中发挥重要作用。因此,通过对松材线虫HAP基因家族鉴定和表达分析,为深入解析松材线虫与寄主间复杂的相互关系提供了新思路,为探索松材线虫病发生机制提供新方向。 To understand the role of histidine acid phosphatase(HAP)secreted in the body of Bursaphelenchus xylophilus,in the process of infesting hosts,the members of HAP gene family of B.xylophilus were identified from bioinformatics perspective by whole genome data of and analyzed for physicochemical properties,signal peptides,transmembrane structural domains,phylogeny,chromosome localization,gene structure and conserved motifs.And the transcriptome data of B.xylophilus inoculated with Pinus thunbergia at different periods and after being stressed by pine metabolitesβ-pinene were used to analyze the gene expression profiles of HAP gene family members,and then to quantify the expression of BxHAP genes in B.xylophilus under different pine metabolite treatments.The results showed that most of the 34 identified members of the HAP gene family were acidic proteins and 12 members were typical effectors.The 34 members belong to six different subfamilies,evenly distributed on the first six chromosomes of B.xylophilus,and the gene structure and conserved motifs remained relatively consistent within each subfamily;the BxHAP genes played important functions in the process of B.xylophilus infestation and had the ability to influence pine metabolites,among which BxHAP5 played an important role in the resistance of B.xylophilus to pine metabolites.Therefore,the identification and expression analysis of the HAP gene family would provide new ideas for the in-depth analysis of the complex interrelationship between B.xylophilus and their hosts,and provide new directions for exploring the mechanism of pine wilt disease occurrence.
