鹿角藤茎叶化学成分研究

摘要 目的:对夹竹桃科鹿角藤属植物鹿角藤 Chonemorpha eriostylis Pitard 的茎叶地上部分进行化学成分研究。方法:采用大孔树脂、硅胶、中压色谱分离凝胶 (MCI)、凝胶及高效液相色谱等方法对鹿角藤茎叶的 95% 乙醇提取物进行分离纯化,并通过一维核磁共振、质谱技术对化合物的结构进行鉴定。用 1,1-二苯基-2-三硝基苯肼(DPPH) 自由基清除法测试分离纯化到的化合物的抗氧化活性。结果:从鹿角藤的 95% 乙醇提取物中分离得到 7 个化合物,分别鉴定为(+)-丁香脂素(1)、异东莨菪内酯(2)、东莨菪内酯(3)、雄甾-1,4-二烯-3,17-二酮 (4)、丝胶树碱 C (5)、清香桂碱 D (6)、鹿角藤碱 (7)。结论:化合物 2、4~6 为首次在该属植物中分离得到。化合物 1 具有较好的抗氧化活性,其半数抑制浓度 (IC) 值为 33. 1 μmol·L。 Objective: To explore the chemical constituents in the stems and leaves of Chonemorpha eriostylis Pitard.Methods: Column chromatograph with macroporous adsorbent resin, silica gel, middle chromatogram isolated gel (MCI),and gel and high performance liquid chromatography (HPLC) were employed to separate and purify 95% ethanol extracts of the stem and leaf extracts of C. eriostylis. The chemical structures of the compounds were identified by nuclear magnetic resonance (NMR) and mass spectrometry (MS). The antioxidant activity of the isolated compounds was tested by the DPPH free radical scavenging assay. Results: Seven compounds were isolated from95% ethanol extracts of stems and leaves of C.eriostylis and identified as (+ ) -syringaresinol (1), isoscopoletin (2), scopoletin (3), androst-1, 4-dien-3, 17-dione (4),funtumafrine C (5), sarcorucinine D (6), and chonemorphine (7). Conclusion: Compounds 2, 4-6 were isolated from Chonemorpha for the first time. Compound 1 has good antioxidant activity, with the ICvalue of 33.1 μmol·L.
