摘要 目的 探索黄芩素抗新型冠状病毒(奥密克戎毒株)的药理活性。方法 利用Autodock Vina软件将黄芩素对接到新型冠状病毒RNA聚合酶晶体结构7BV2的底物结合位点处,研究二者分子间的相互作用。而后利用奥密克戎毒株转染的Vero细胞模型测试黄芩素的抗病毒活性。结果 黄芩素可较好的结合于靶酶7VB2的底物进入通道处从而阻碍正常核苷酸类底物的进入,对靶酶执行的核酸复制生物学功能起到抑制作用;细胞水平的药理活性测试发现黄芩素具有抑制奥密克戎毒株核酸复制的作用。结论 黄芩素具有抗新型冠状病毒及其变异株的药理活性,可为临床治疗方案制定提供基础实验性的支持。 Objective To explore the pharmacological activity of baicalin against SARS-CoV-2(Omicron subvariant).Methods Baicalin was docked to the substrate binding site of SARS-CoV-2 RNA polymerase crystal structure 7 VB2 using Autodock Vina software to study the intermolecular interaction between them.The antiviral activity of baicalin was then tested using Vero cell model transfected with Omicron subvariant.Results Baicalein can bind well to the substrate entry channel of the target enzyme 7 BV2,and thus blocking the entry of normal nucleotide substrates,and inhibiting the biological function of nucleic acid replication performed by the target enzyme.The pharmacological activity test at the cellular level revealed that baicalin had the effect of inhibiting the nucleic acid replication of Omicron subvariant.Conclusion Baicalin has pharmacological activity against SARS-CoV-2 and its subvariant,which can provide basic experimental support for the development of clinical treatment protocols.
