摘要
目的 采用超高效液相色谱-四级杆-飞行时间质谱(UPLC-Q-TOF/MS)法分析红豆蔻醇提物的主要化学成分及胃溃疡寒证大鼠口服后的入血成分。方法 回流提取法提取药物;大鼠灌胃醇提物后腹主动脉取血;分析采用Acquity HSS T3 C_(18)色谱柱(100 mm×2.1 mm,1.8μm);流动相0.1%甲酸水-甲醇;梯度洗脱,流速0.4 mL·min^(-1);柱温35℃,MSE扫描正负离子模式检测。结果 综合色谱-质谱检测信息,包括色谱保留时间、分子量、离子碎片,结合相关文献报道,从红豆蔻乙醇提取物中鉴定出14个化学成分;胃溃疡寒证大鼠口服红豆蔻醇提物后,血清样本共鉴定出5个入血成分。结论 上述研究结果初步阐释了红豆蔻的药效物质基础,为研究红豆蔻抗胃溃疡寒证的药效物质提供了一定依据。
Objective The chemical constituents of Hongdoukou(Galangae Fructus) alcohol extract and the blood components of rats with gastric ulcer cold syndrome after oral administration were analyzed by UPLC-Q-TOF/MS.Methods The reflux extraction method was used to extract the drugs.The blood was collected from the abdominal aorta of rats after intragastric administration of alcohol extract.Acquity HSS T3 C_(18) column(100 mm×2.1 mm,1.8 μm) was used for analysis.The mobile phase was 0.1% formic acid water-methanol.The gradient elution rate was 0.4 mL·min^(-1).The column temperature was 35 ℃,and positive and negative ion modes were detected by MSE scanning.Results Based on the chromatography-mass spectrometry analysis,including chromatographic retention time,molecular weight,ion fragments and related literature reports,14 chemical constituents were identified from the ethanol extract of Hongdoukou(Galangae Fructus).After oral administration of alcohol extract of Hongdoukou(Galangae Fructus) in rats with gastric ulcer and cold syndrome,a total of 5 components were identified in serum samples.Conclusion The above research results preliminarily explained the pharmacological substance basis of Hongdoukou(Galangae Fructus) and provided a certain basis for the study of the pharmacological substance of Hongdoukou(Galangae Fructus) against cold syndrome of gastric ulcer.
