CⅡTA基因沉默小鼠鼻息肉组织中胶原纤维沉积和炎症反应的初步研究

摘要 目的:探究CⅡTA在慢性鼻窦炎伴鼻息肉(chronic rhinosinusitis with nasal polyps,CRSwNP)发病中对胶原纤维沉积和炎症反应的可能作用及机制。方法:构建野生型鼻息肉小鼠模型和CⅡTA^(-/-)小鼠鼻息肉模型,选取小鼠鼻腔外侧壁组织,通过HE染色法观察各组小鼠鼻黏膜组织的形态学结构及炎性细胞浸润情况。通过Masson三色胶原染色法观察细胞外基质中胶原沉积情况,ELISA检测鼻腔灌洗液中碱性成纤维细胞生长因子(bFGF)及炎症因子(TNF-α、IL-6和IL-17)的表达情况。结果:NP小鼠组黏膜下炎性细胞浸润增多、胶原沉积增加,灌洗液中炎性因子浓度升高鼻腔。同WT+NP组小鼠相比,CⅡTA^(-/-)+NP小鼠的黏膜病理学变化减轻,炎性因子浓度降低。结论:小鼠不表达CⅡTA可能通过减少黏膜下胶原纤维沉积和减轻炎症反应的方式延缓鼻息肉的发展。 Objective:To investigate the possible role and mechanism of CⅡTA in collagen fibril deposition and inflammatory response in sinusitis with nasal polyp.Methods:Establish the model of wild type NASAL POLYP in mice and CⅡTA^(-/-)model of nasal polyp in mice,the tissue of lateral wall of nasal cavity was selected,and the morphological structure and inflammatory cell infiltration of nasal mucosa were observed by HE staining.The collagen deposition in the extracellular matrix was observed by masson’s three-color collagen staining.The expression of basic fibroblast growth factor and inflammatory factors(TNF-Α,IL-6 and IL-17)in nasal lavage fluid was detected by elisa.Results:In NP group,submucosal inflammatory cell infiltration and collagen deposition were increased,and the concentration of inflammatory factor in lavage solution was increased.Compared with WT+NP group,CⅡTA^(-/-)+NP group showed reduced mucosal pathological changes and inflammatory factor concentration.Conclusion:The lack of CⅡTA expression in mice may delay the development of nasal polyps by reducing submucosal collagen deposition and reducing inflammatory response.
