剔除目标成分的丹参注射液对大鼠嗜碱性粒细胞白血病细胞和大鼠过敏反应的影响

摘要 目的 探讨剔除目标成分后的丹参注射液对大鼠嗜碱性粒细胞白血病(RBL-2H3)细胞和大鼠过敏反应的影响。方法 选取RBL-2H3细胞,分别以25、50、100μg/ml的丹参和剔除目标成分后的丹参注射液处理24小时后,比较各组细胞脱颗粒、β-氨基己糖苷酶含量、组胺含量及蛋白(p-p38、p-cJNU、p-Syk)表达。将雄性SD大鼠随机分为丹参注射液组、剔除目标成分后注射液组、正常组和阳性对照组(天花粉蛋白)各8只,按照临床等倍剂量分别腹腔注射丹参注射液组、剔除目标成分后注射液组、生理盐水及天花粉蛋白。比较各组大鼠血清被动阿瑟斯反应、肺组织病理结构、组胺含量和总IgE浓度。结果 在RBL-2H3细胞中,丹参和剔除目标成分后的丹参注射液干预组RBL-2H3细胞的脱颗粒率、β-氨基己糖苷酶释放率、组胺含量和p-p38、p-cJNU和p-Syk蛋白的表达显著高于阴性对照组;相同浓度的丹参和剔除目标成分后的丹参注射液干预下,剔除目标成分后的丹参注射液组细胞中的这些指标显著低于丹参组,差异有统计学意义(P<0.05)。在SD大鼠中,丹参和剔除目标成分后的丹参注射液干预组SD大鼠被动阿瑟斯反应、肺组织病理损伤、SD大鼠血清和肺组织中组胺含量和总IgE浓度显著高于正常组;剔除目标成分后的丹参注射组显低于丹参注射组,差异有统计学意义(P<0.05)。结论 相较于丹参注射液,剔除目标成分后的丹参注射液可减弱RBL-2H3细胞和大鼠过敏反应。 Objective To investigate the effect of Salvia miltiorrhiza injection after removal of target ingredients on the allergic reactions of rats and RBL-2 H3 cells.Methods RBL-2 H3 cells were selected and treated with different concentrations of 25, 50 and 100 μg/ml of Salvia miltiorrhiza or Salvia miltiorrhiza after removal of target ingredients for 24 hours. Cell degranulation, β-hexosaminidase content, histamine content, and expressions of proteins of p-p38, p-cJNU and p-Syk were compared among various dose groups. Male SD rats were randomly divided into a Salvia miltiorrhiza injection group, a Salvia miltiorrhiza injection after removal of target ingredients group, normal group and positive control group(trichosanthin), 8 in each group. According to the clinical equivalent dose, the groups were injected with Salvia miltiorrhiza, Salvia miltiorrhiza after removal of target ingredients, normal saline and trichosanthin. Passive Arthurs reaction, pathological structure of lung tissue, histamine content and total IgE concentration in lung tissue or serum were compared among the groups.Results After treatment with Salvia miltiorrhiza or Salvia miltiorrhiza after removal of target ingredients, the degranulation rate, β-hexosaminidase release rate, histamine content and protein expressions of p-p38, p-cJNU and p-Syk in RBL-2 H3 cells were significantly higher than those in the negative control group. The indexes in the cells treated with Salvia miltiorrhiza after removal of target ingredients were significantly lower than those in the cells treated with Salvia miltiorrhiza at the same doses(P<0.05). After treatment of Salvia miltiorrhiza and Salvia miltiorrhiza after removal of target ingredient, the passive Arthritis response, lung histopathological damage, histamine content and total IgE concentration in serum or lung tissue in SD rats were significantly higher than those in the normal group, and those in the Salvia miltiorrhiza after removal of target ingredients group was lower than those in the Salvia miltiorrhiza group(P<0.05).Conclusion Compared with Salvia miltiorrhiza injection, Salvia miltiorrhiza injection after removal of target ingredients can attenuate the allergic reactions in RBL-2 H3 cells and SD rats.
