胃复春胶囊提取物对5-氟尿嘧啶诱导人胃癌细胞凋亡的影响

The Promoting Effect of Weifuchun Capsule Extract on Human Gastric Cancer Cells Apoptosis Induced by 5-fluorouracil

目的探讨胃复春胶囊提取物对5-氟尿嘧啶干预人胃癌细胞氧化还原水平和诱导细胞凋亡作用的影响。方法人胃癌细胞株MGC-803用5-氟尿嘧啶处理组后,受试组同时添加终浓度为分别为50、100、200μg/ml的胃复春胶囊提取物,对照组添加相同体积的DMSO溶液,培养24 h,采用MTT法测定细胞活力;采用流式细胞术Annexin V/PI双染色检测细胞凋亡;Western blot法检测与细胞凋亡相关的蛋白表达情况;测定各组细胞氧化水平SOD、MDA和GSH-Px参数,评价抗氧化能力。结果5-氟尿嘧啶可显著降低人胃癌MGC-803细胞活力,诱导细胞凋亡,并使SOD和GSH-Px活性降低,MDA含量增加;5-氟尿嘧啶处理细胞的同时添加胃复春胶囊提取物后,MGC-803细胞相对活力显著下降,细胞凋亡增强,胞内SOD和GSH-Px活性进一步降低,MDA含量增加;Western Blot结果显示经过5-氟尿嘧啶处理,MGC-803细胞中促凋亡蛋白Bax表达量显著上升,抗凋亡蛋白Bcl-2表达量降低;胃复春胶囊提取物可以显著上调Bax表达量,下调Bcl-2表达量。结论胃复春胶囊提取物可明显改变人胃癌细胞中凋亡及抗氧化能力,对5-氟尿嘧啶诱导的细胞凋亡起到促进作用。

Objective To investigate the effect of Weifuchun capsule extract on the redox level and apoptosis of human gastric cancer cells treated with 5-fluorouracil.Methods Human gastric cancer cell line MGC-803 was treated with 5-fluorouracil for 24 hours,and the extract of Weifuchun capsule with final concentration of 50,100 and 200μg/ml was added to the test group,while the same volume of DMSO solution was added to the control group.The expression levels of apoptosis related proteins were detected by western blot,and the oxidative levels of SOD,MDA and GSH Px parameters were measured to evaluate the antioxidant capacity.Results 5-fluorouracil could significantly reduce the activity of MGC-803 cells,induce apoptosis,decrease the activities of SOD and GSH Px,and increase the content of MDA.After5-fluorouracil treatment and addition of Weifuchun capsule extract,the relative activity of MGC-803 cells decreased significantly,the apoptosis of MGC-803 cells increased,the activities of SOD and GSH-Px further decreased,and the content of MDA increased.After 5-fluorouracil treatment,the expression of pro-apoptotic protein Bax in MGC-803 cells increased significantly,while the expression of antiapoptotic protein Bcl-2 decreased.Weifuchun capsule extract could significantly up regulate the expression of Bax and down regulate the expression of Bcl-2.Conclusion Weifuchun capsule extract can significantly change the apoptosis and antioxidant capacity of human gastric cancer cells,and promote the apoptosis induced by 5-fluorouracil.
