基于免疫亲和柱真菌毒素全自动亲和纯化仪净化方法适用性评价

摘要 分别采用自主研发的全自动亲和纯化仪和人工免疫亲和柱对小麦、玉米、花生、饲料等样品进行前处理,并应用高效液相色谱法测定样品中的毒素含量,同时对2种前处理方法的操作时间、洗脱液体积、加标回收率进行比较。结果表明:当样品数量较少(1~8个)时,2种前处理方法所用时间区别不大;当样品数量大于8时,全自动亲和纯化仪的用时更短。2种前处理方法所用洗脱液体积差异不大。仪器的加标平均回收率在87.20%~105.50%之间,变异系数小于3.41%。手工操作的加标平均回收率为81.00%~106.00%,回收率变异系数为7.07%。在实际阳性样品检测中,仪器的检测结果稳定性优越,这表明全自动免疫亲和纯化仪可在食品安全检测领域得以应用。 The samples of wheat,corn,peanut and feed were pretreated by self-developed automatic affinity purifier and manual immunoaffinity column,respectively.The mycotoxins content in the samples was determined by HPLC.The operation time,eluent volume and recovery rate of the two pretreatment methods were compared.The results showed that when the number of samples was 1-8,there was no significant difference in the time between the two pretreatment methods.When the number of samples was more than 8,the time of automatic affinity purifier was shorter.There was no significant difference in eluent volume between the two methods.The average recovery rate of the instrument was between 87.20 % and 105.50 %,and the coefficient of variation was less than 3.41 %.The average recovery rate of manual operation was 81.00 %-106.00 %,and the coefficient of variation was 7.07 %.In the actual positive sample detection,the stability of the instrument was superior,indicating the automatic immunoaffinity purifier could be applied in the field of food safety detection.
