摘要
目的探讨代谢综合征(MS)痰证形成的机制及病理基础。方法50只大鼠随机分为空白组12只和造模组38只。造模组大鼠运用高糖高脂高盐饲养法建立MS痰证大鼠模型。共24只大鼠造模成功,随机分为模型组和二陈汤组各12只。二陈汤组予以二陈汤5.3 g/kg灌胃,模型组及空白组予以生理盐水10 ml/kg灌胃,均每日1次,连续4周。采用分光光度法分别于造模0周、4周、8周、12周检测空白组与造模组血浆乳酸含量。给药后空白组、模型组、二陈汤组采用ELISA法检测血清及肝组织三磷酸腺苷(ATP)水平,采用ELISA法及Real-Time PCR检测肝组织柠檬酸合成酶(CS)、肉毒碱脂酰转移酶I(CPT-1)、乳酸脱氢酶(LDH)、低氧诱导因子1α(HIF-1α)水平及mRNA表达情况。结果造模期间,空白组与造模组乳酸水平比较差异具有统计学意义,造模第8周和第12周,造模组大鼠乳酸含量较空白组升高(P<0.05)。给药后,与空白组比较,模型组和二陈汤组大鼠血清及肝组织ATP的含量升高(P<0.05),模型组大鼠肝组织LDH、CS、CPT-1、HIF-1α水平均升高(P<0.05);与模型组比较,二陈汤组大鼠血清及肝组织ATP的含量,肝CPT-1 mRNA表达及肝LDH、CPT-1、HIF-1α水平均明显降低(P<0.05)。结论能量失衡促进MS痰证发生发展,能量代谢途径改变是MS痰证形成的病理基础。
Objective To explore the mechanism and pathological basis of phlegm syndrome in metabolic syn⁃drome(MS).Methods Fifty rats were randomly divided into blank group(12 rats)and modeling group(38 rats).The rat model of MS with phlegm syndrome was established by feeding high sugar,high fat and high salt diet.Twentyfour successfully modeled rats were randomly divided into a model group and a Erchen Decoction(二陈汤)group,with 12 rats in each group.The Erchen Decoction group was given 5.3 g/kg of Erchen Decoction by gavage,while the other groups were administered with 10 ml/kg of normal saline by gavage,once daily for four weeks.Plasma lactic acid levels in the blank group and the model group were detected by spectrophotometry at 0,4,8 and 12 weeks,respec⁃tively.Adenosine triphosphate(ATP)levels in serum and liver tissues of the three groups were detected by ELISA.Citric acid synthase(CS),carnitine palmitoyltransferase I(CPT-1),lactate dehydrogenase(LDH),hypoxia inducible factor-1α(HIF-1)levels and mRNA expression in liver tissue were detected by ELISA and real-time PCR.Results During the modeling period,the difference of lactic acid level between the blank group and the model group was statistically significant.At 8 and 12 weeks of modeling,the lactic acid level of the model group was higher than that of the blank group(P<0.05).At the end of the 16th week during the experiment,compared with those of the blank group,the ATP content in serum and liver tissue of the model group and the Erchen Decoction group increased(P<0.05),and the levels of LDH,CS,CPT-1 and HIF-1αin the liver tissue of the model group increased(P<0.05).Compared with those of the model group,the contents of ATP in serum and liver tissue,the expression of CPT-1 mRNA in liver and the levels of LDH,CPT-1 and HIF-1αin liver tissue of the Erchen Decoction group signifi⁃cantly decreased(P<0.05).Conclusion The energy imbalance promotes the development of MS phlegm syn⁃drome,and the change in the energy metabolic pathway is the pathological basis.
