4种自粘接树脂水门汀对牙周膜成纤维细胞增殖的影响 被引量:2

摘要 目的比较4种自粘接树脂水门汀对人牙周膜成纤维细胞(HPDLFs)增殖的影响。方法将4种自粘接树脂水门汀试件(RelyX Unicem、SAC、Duo-Link和PermaCem)分别浸泡1、3、5、7 d,取浸提液培养HPDLFs 48 h,采用CCK-8法测定培养液的光密度(OD),计算细胞相对增殖率(RGR),并结合镜下细胞形态学分析。比较分析在4种自粘接树脂水门汀浸提液的作用下HPDLFs的RGR差异。结果4种自粘接树脂水门汀1 d的浸提液对HPDLFs增殖均无抑制作用,毒性分级为0级。浸提3 d时,Unicem组、SAC组和Duo-Link组抑制细胞增殖,表现为轻中度细胞毒性,其中Unicem组毒性分级为3级、SAC组和Duo-Link组毒性分级为2级。浸提5 d时,除了SAC组为毒性2级,其余3组对细胞均无毒性。光镜下观察细胞形态未见明显异常。随浸泡时间的延长,毒性分级逐渐降低,浸提7 d的树脂水门汀均无细胞毒性。PermaCem组浸泡1、3、5、7 d均对细胞无毒性。结论4种自粘接树脂水门汀浸提液对HPDLFs增殖的抑制作用均表现为3 d达到最强,7 d趋于无细胞毒性、细胞增殖恢复正常。4种水门汀均是良好的可供选择的粘接剂,而PermaCem的生物相容性更好些。 Objective To compare the effects of four kinds of self-adhesive resin cements on the proliferation of human periodontal ligament fibroblasts(HPDLFs).Methods Four kinds of self-adhesive resin cements(RelyX Unicem,SAC,Duo-Link,PermaCem)were extracted for 1,3,5 and 7 days.The extracts were taken to culture HPDLFs for 48 hours.The optical density(OD)of the culture solution was measured by CCK-8 method to calculate the relative growth rate(RGR).The differences in HPDLCs RGR were compared and analyzed under the action of the extracts of four self-adhesive resin cements.Results Four kinds of self-adhesive resin cement extract for 1 day had no inhibitory effect on the proliferation of HPDLFs and the toxicity was classified as 0.After 3 days of leaching,Unicem,SAC and Duo-Link groups inhibited cell proliferation,showing mild to moderate cytotoxicity.Among them,the toxicity classification of Unicem group was grade 3,and the toxicity classification of SAC and Duo-Link group was grade 2.After 5 days of extracting,the other three groups had no toxicity to cells except the SAC group,which was of class 2 toxicity.No obvious abnormality was found in cell morphology under light microscope.With the prolongation of extracting time,the toxicity grading gradually decreased,and no cytotoxicity was found in the resin cement after 7 days of extracting.PermaCem had no toxicity to cells regardless of extracting for 1,3,5 and 7 days.Conclusion The inhibitory effect of the self-adhesive resin cement extract on the proliferation of HPDLFs is the strongest after 3 days of extracting,and tends to have no cytotoxicity after 7 days of extracting,and cell proliferation returns to normal.Four kinds of self-adhesive resin cements are good adhesives to choose from.By comparison,PermaCem has better biocompatibility.
