基于生物信息学方法分析HMMR在结直肠癌中的表达及临床意义

摘要 目的探究透明质酸介导的运动受体(HMMR)基因在结直肠癌(CRC)中的表达及临床意义。方法在本研究中,使用Oncomine、GEPIA、BioGPS数据库研究HMMR在结直肠癌组织和正常结直肠组织中表达的差异;使用Ualcan数据库分析HMMR与结直肠癌临床病理特征的关系;使用GEPIA数据库及COX分析研究HMMR表达水平对CRC患者预后的影响;使用String数据库绘制HMMR蛋白互作网络图及GO功能注释和KEGG通路富集分析。使用TISIDB数据库分析HMMR在免疫浸润中的表达及不同分子亚型和免疫亚型中表达的差异。结果通过Oncomine、GEPIA、BioGPS分析:与正常组织相比,HMMR在结直肠癌中表达水平明显升高(P<0.05)。通过HPA数据库显示,免疫组化结果提示HMMR在结直肠癌中染色成阳性。通过GEPIA数据库发现,HMMR高表达的CRC预后比HMMR低表达的患者预后更好(P=0.02)。通过COX分析发现,HMMR基因是结直肠癌患者总体生存(OS)的良好预后因子(P=0.002)。通过PPI蛋白互作、GO富集、KEGG通路收集到与HMMR相关蛋白18组。主要富集于10种细胞学组分、5种分子功能和11种生物学过程,并参与了4条信号通路。通过TISIDB数据库分析HMMR mRNA与NK细胞、B细胞、巨噬细胞、CD4+T细胞和肥大细胞等细胞在结直肠癌组织的免疫微环境中的表达明显相关。结论HMMR在结直肠癌中呈高表达,HMMR的表达可协助评估结直肠癌患者的预后情况。 【Objective】To investigate the expression and clinical significance of hyaluronic acid mediated receptor(HMMR)gene in colorectal cancer(CRC).【Methods】In this study,Oncomine,GEPIA and BioGPS databases were used to study the difference of HMMR expression between colorectal cancer tissues and normal colorectal tissues.Ualcan database was used to analyze the relationship between HMMR and clinicopathological features of colorectal cancer.GEPIA database and COX analysis were used to investigate the effect of HMMR expression level on prognosis of CRC patients.String database was used to draw HMMR protein interaction network diagram,GO functional annotation and KEGG pathway enrichment analysis.TISIDB database was used to analyze the expression of HMMR in immune infiltration and the difference of expression among different molecular subtypes and immune subtypes.【Results】By Oncomine,GEPIA and BioGPS analysis,the expression level of HMMR in colorectal cancer was significantly higher than that in normal tissues(P<0.05).HPA database showed that immunohistochemical results indicated that HMMR was positive in colorectal cancer.The GEPIA database showed that CRC with high HMMR expression had a better prognosis than those with low HMMR expression(P=0.02).COX analysis showed that HMMR gene was a good prognostic factor for overall survival(OS)in colorectal cancer patients(P=0.002).Eighteen groups of HMMR-related proteins were collected through PPI protein interaction,GO enrichment and KEGG pathway.They are enriched in 10 cytological components,5molecular functions and 11 biological processes,and participate in 4 signaling pathways.TISIDB database analysis showed that HMMR mRNA was significantly correlated with the expression of NK cells,B cells,macrophages,CD4+T cells and mast cells in the immune microenvironment of colorectal cancer tissues.【Conclusion】HMMR is highly expressed in colorectal cancer,and the expression of HMMR can help evaluate the prognosis of colorectal cancer patients.
