大球盖菇种质资源遗传多样性 被引量:1

Mycelial Morphology and Genetic Diversities of Germplasm Resources in Stropharia rugoso-annulata

摘要 对收集到的10株大球盖菇进行聚类并进行亲缘关系的分析。方法:应用同工酶和RAPD技术对从美国、波兰和中国收集的10株大球盖菇菌株的遗传多样性进行了分析。结果:同工酶分析结果表明,大球盖菇菌株间遗传差异较大,最大的两类之间相似系数仅为0.0096,有5株相似系数在0.5000以下,St2、St4、St5间相似系数在0.5以上,特别是St4和St5相似系数为1.0000。RAPD分析结果显示,20个随机引物中有16个随机引物的扩增效果较好,分别扩增出8~21个条带,共扩增出241条条带,片段大小在200~4000 bp之间。结论:这些菌株之间的相似系数用UPGMA进行聚类分析可以看出,St2、St4、St5相似系数为0.8248聚为一类,说明它们之间的遗传关系较近,进一步从核酸水平验证了同工酶实验结果。本研究说明大球盖菇菌株间具有丰富的遗传多样性。 Objective:Relationship and cluster analysis is done based on similarity coefficient of 10 Stropharia rugoso-annulata isolates.Methods:Ten isolates of Stropharia rugoso-annulata collected from America,Poland and China were studied by isoenzyme and RAPD technology in this research.Results:The dendrogram of cluster analysis based on the PAGE of esterase isoenzyme showed that the genetic difference between the isolates was big,and the similarity coefficient of the two biggest groups was only 0.0096.The similarity coefficients of five isolates were below 0.5000.The similarity coefficient of St2,St4 and St5 was above 0.5000.Moreover,St4 and St5 were almost the same since the coefficient was 1.0000.The results of RAPD by 20 primers showed that the amplification effect of 16 random primers was better,with 8 to 21 amplification bands,respectively.The total bands were 241,and the fragments were from 200 bp to 4000 bp.Conclusion:The results of dendrogram of cluster analysis based on DNA similarity coefficient by using UPGMA demonstrated that St2,St4 and St5 belonged to one group and the similarity coefficient was 0.8248.This result showed their genetic relationship was relatively close,which was further verified in the results of isoenzyme from DNA level.Therefore,the genetic diversity of Stropharia rugoso-annulata was very abundant.
