硫酸氨基葡萄糖增强小鼠免疫功能的研究

摘要 目的 研究硫酸氨基葡萄糖对小鼠免疫功能的增强作用。方法 将160只SPF级雌性昆明种小鼠随机分为对照组和低、中、高3个剂量组,分别相当于0、0.142、0.285和0.850 g/kg硫酸氨基葡萄糖,每组40只,经口灌胃给予不同浓度的硫酸氨基葡萄糖水溶液,灌胃容积为20 mL/kg,每日1次,连续30 d,末次灌胃24 h后分别进行脾淋巴细胞转化试验、迟发型变态反应试验、溶血空斑试验、血清溶血素的测定、小鼠碳廓清试验、巨噬细胞吞噬鸡红细胞试验及自然杀伤细胞活性试验。结果 各组动物生长活动良好,各剂量硫酸氨基葡萄糖对小鼠体质量及免疫器官脏器系数无影响。0.850 g/kg组小鼠半数溶血值为60.2,抗体生成细胞数为7.9×10^(3)/全脾,均高于对照组(半数溶血值为50.2,抗体生成细胞数为5.4×10^(3)/全脾),差异均有统计学意义(均P<0.05);小鼠脾淋巴细胞转化试验中,0.285和0.850 g/kg组细胞吸光度分别为0.325和0.310,均高于对照组(0.275),差异均有统计学意义(均P<0.05);0.285和0.850 g/kg组小鼠自然杀伤细胞活性分别为98.7%和97.7%,均高于对照组(68.8%),差异均有统计学意义(均P<0.05)。硫酸氨基葡萄糖对小鼠迟发性变态反应、腹腔巨噬细胞吞噬鸡红细胞的吞噬率和碳廓清吞噬指数均无影响。结论 硫酸氨基葡萄糖具有良好的增强小鼠免疫功能的作用,其作用主要表现为可提高体液免疫、细胞免疫和自然杀伤细胞活性。 Objective To study the effect of glucosamine sulfate on enhancing immunological function of mice. Methods 160 SPF female Kunming mice were randomly divided into the control group and low,medium and high dose groups,which was equivalent to 0,0.142,0.285 and 0.850 g/kg glucosamine sulfate,respectively,40 animals in each group. They were orally given glucosamine sulfate solution with different concentrations once a day for 30 days,and the gavage volume was 20 mL/kg. The spleen lymphocyte transformation test,delayed type hypersensitivity test,antibody cell generation test,serum hemolysin level determination,mouse carbon clearance ability test,phagocytosis of chicken red blood cells by macrophages test and natural killer(NK) cell activity test were performed 24 h after the last gavage. Results The animals in each group grew well,and each dose of glucosamine sulfate had no effect on the body weight and organ coefficient of immune organs. In 0.850 g/kg group,the half hemolysis value was 60.2,and the number of antibody producing cells was 7.9×10^(3)/spleen,which were higher than those in control group(50.2 and 5.4×10^(3)/spleen),and the differences were statistically significant(all P<0.05). In the transformation of spleen lymphocytes,the cell absorbance in0.285 and 0.850 g/kg groups was 0.325 and 0.310 respectively and both higher than that in control group(0.275),and the differences were statistically significant(all P<0.05). The NK cell activity in 0.285 and 0.850 g/kg groups was 98.7% and 97.7%respectively,higher than that in the control group(68.8%),and the difference was statistically significant(all P <0.05). The glucosamine sulfate had no effect on delayed allergic reaction in mice,phagocytic rate of peritoneal macrophages in chicken red blood cells and carbon clearance phagocytic index. Conclusion The glucosamine sulfate can enhance the immune function of mice,mainly in enhancing humoral immunity,cellular immunity and NK cell activity.
