摘要
目的 研究CD3、Ki-67和Bcl-2蛋白在不同宫颈病变中的表达水平以预测临床结果,并研究这些蛋白与临床病理特征之间的关系。方法 收集27例正常宫颈组织标本,12例宫颈上皮内瘤变Ⅰ(CINI)标本,28例宫颈上皮内瘤变Ⅱ~Ⅲ(CINⅡ~Ⅲ)标本,51例宫颈癌标本。使用半定量免疫组织化学方法检测组织样本中CD3、Ki-67和Bcl-2的表达水平,分析他们之间的相关性以及宫颈癌患者的临床病理特征与CD3、Ki-67、Bcl-2表达之间的关系。结果 从CINⅠ~Ⅲ到宫颈癌,Ki-67和Bcl-2的表达水平升高,而CD3的表达顺序下降。CD3的阳性表达率与FIGO分期和淋巴结转移相关,而Ki-67的表达仅与肿瘤分化状态相关。Bcl-2的表达分别与FIGO分期、肿瘤分化状态和淋巴结转移相关。Spearman秩相关检验显示,CD3与Ki-67和Bcl-2的表达均呈负相关关系。结论 CD3、Ki-67、Bcl-2蛋白参与了宫颈上皮内瘤变和宫颈癌的发生发展,其检测有助于宫颈上皮内瘤变和宫颈癌的早期检测和分级。
Objective The expression levels of CD3, Ki-67 and Bcl-2 proteins in different cervical lesions to predict clinical outcomes were studied, the relationship between these proteins and clinicopathological features was also investigated. Methods 27 normal cervix tissue samples, 12 cervical intraepithelial neoplasia Ⅰ(CINⅠ) samples, 28 CINⅡ-Ⅲ samples, and 51 cervical cancer samples were collected. We detected the expression levels of CD3, Ki-67, and Bcl-2 in these tissue samples using semiquantitative immunohistochemistry, and analyzed correlation among the expression levels of these three proteins. The relationships between clinicopathological features and CD3, Ki-67 and Bcl-2 expressions in patients with cervical cancer were investigated as well. Results The expression levels of Ki-67 and Bcl-2 were increased from normal cervix tissue to CINⅠ-Ⅲ to cervical cancer, while CD3 expression was declined in the same order. Moreover, positive expression rate of CD3 was correlated with FIGO stage and lymph node metastasis, while Ki-67 expression with only tumor differentiation status. In addition, Bcl-2 expression was associated with FIGO stage,tumor differentiation status, and lymph node metastasis, respectively. Spearman rank correlation test showed that CD3 was negatively correlated to Ki-67 and Bcl-2 expressions, respectively. Conclusions CD3, Ki-67and Bcl-2 proteins were involved in the occurrence and development of CIN and cervical cancer, and detection of these proteins could assist the early detection and grading of CIN and cervical cancer.
