摘要
目的:探究毛水苏多糖(polysaccharides from Stachys baicalensis,SBP)对糖尿病小鼠肠道菌群的影响。方法:采用高糖饲料喂养结合腹腔注射链脲佐菌素(STZ)诱导建立Ⅱ型糖尿病模型,连续灌胃28d后,考察毛水苏多糖干预后回肠组织显微结构的变化,通过GC-MS法测定短链脂肪酸分泌含量,并结合Illumina NovaSeq高通量测序技术对肠道菌群组成及多样性进行分析。结果:低剂量毛水苏多糖可明显改善小鼠回肠组织显微结构,促进肠道内乙酸、丙酸及丁酸含量水平的恢复,使厚壁菌门、类杆菌门菌群丰度得以恢复。结论:低剂量毛水苏多糖能够改善STZ诱导的糖尿病小鼠肠道菌群紊乱,其作用机制可能与调控产酸菌的丰度情况,促进短链脂肪酸的含量分泌有关。
Objective:To explore the effect of polysaccharides from Stachys baicalensis(SBP)on the intestinal flora of diabetic mice.Methods:Streptozocin(STZ)induction combined with high-sugar and high-fat feed was used to establish a diabetic mice model.Mice was continuous given intragastric administration for 28 d,the changes in the microstructure of ileum tissue after SBP intervention were investigated,the secretion of short-chain fatty acids was determined by GC-MS,and the composition and diversity of intestinal flora were analyzed by Illumina Nova Seq high-throughput sequencing technology.Results:Low-dose SBP can significantly improve the microstructure of ileum tissue,speed up the recovery by acetic acid,propionic acid and butyric acid in the intestine,and restore abundance of Firmicutes and Bacteroidetes.Conclusion:Low-dose SBP can improve the disturbance of intestinal flora in STZ-induced diabetic mice,and its mechanism may be related to regulating the abundance of acid-producing bacteria and promoting the secretion of short-chain fatty acids.
