高效液相色谱法同时测定和营合剂中阿魏酸和丹酚酸B含量

Simultaneous Determination of Ferulic Acid and Salvianolic Acid B in Heying Mixture by HPLC

目的建立同时测定和营合剂中阿魏酸和丹酚酸B含量的高效液相色谱法。方法色谱柱为Waters XBridge C18柱(250 mm×4.6 mm,5μm);流动相为乙腈(A)-0.2%磷酸水溶液(B),梯度洗脱(0~5 min时5%A~10%A,5~15 min时10%A~14%A,15~25 min时14%A~15%A,25~30 min时15%A~17%A,30~45 min时17%A~21%A,45~55 min时21%A~22%A,55~63 min时22%A);流速为1.0 mL/min;柱温为30℃;检测波长为321 nm;进样量为10μL。结果阿魏酸和丹酚酸B的质量浓度分别在2.27~22.74μg/mL(r=0.9999,n=6)和27.05~270.48μg/mL(r=0.9997,n=6)范围内与峰面积线性关系良好;平均加样回收率分别为100.75%和99.06%,RSD分别为1.71%和2.24%(n=6);精密度、稳定性、重复性试验的RSD均小于2.50%(n=6)。每1 mL样品中分别含阿魏酸7.77μg和丹酚酸B 193.17μg(n=3)。结论该方法操作简便、结果准确、稳定性和重复性均良好,可用于和营合剂中阿魏酸和丹酚酸B含量的测定。

Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of ferulic acid and salvianolic acid B in Heying Mixture.Methods The chromatographic column was Waters XBridge C18column(250 mm×4.6 mm,5μm),and the mobile phase was acetonitrile(A)-0.2%phosphoric acid aqueous solution(B)with gradient elution(5%A-10%A at 0-5 min,10%A-14%A at 5-15 min,14%A-15%A at 15-25 min,15%A-17%A at 25-30 min,17%A-21%A at 30-45 min,21%A-22%A at 45-55 min and 22%A at 55-63 min).The flow rate was 1.0 mL/min,the column temperature was 30℃,the detection wavelength was 321 nm,and the injection volume was 10μL.Results The linear ranges of ferulic acid and salvianolic acid B were 2.27-22.74μg/mL(r=0.9999,n=6)and 27.05-270.48μg/mL(r=0.9997,n=6),respectively.The average recoveries of ferulic acid and salvianolic acid B were 100.75%and 99.06%with RSDs of 1.71%and 2.24%(n=6),respectively.The RSDs of precision,stability and repeatability tests were lower than 2.50%(n=6).There were 7.77μg of ferulic acid and 193.17μg of salvianolic acid B each 1 mL of sample(n=3).Conclusion This method is simple,accurate,stable and repeatable,which can be used for the determination of ferulic acid and salvianolic acid B in Heying Mixture.
