基于血清抗体芯片法探讨恶性肿瘤患者表皮生长因子受体抑制剂治疗致皮肤瘙痒的机制

摘要 目的:基于血清抗体芯片法探索恶性肿瘤患者中表皮生长因子受体抑制剂(epidermal growth factor receptor inhibitors, EGFRIs)所致皮肤瘙痒的机制。方法:纳入接受EGFRIs治疗的患者,根据是否出现皮肤瘙痒分为试验组和对照组,采集外周静脉血,通过抗体白芯片对炎症因子的表达进行分析。结果:血清学分析的差异蛋白为粒细胞巨噬细胞刺激因子(granulocyte-macrophage colony-stimulatingfactor, GM-CSF)、IL-16、巨噬细胞刺激因子(macrophage colony-stimulating factor,MCSF),通路富集显示巨噬细胞刺激因子相关的信号通路可能参与EGFRIs所致皮肤瘙痒的发生。结论:GM-CSF诱导的巨噬细胞炎症反应可能参与EGFRIs所致皮肤瘙痒的发生。 Objective:To reveal the mechanism of pruritus induced by epidermal growth factor receptor inhibitors(EGFRIS) based on serological analysis in patients with malignant tumors.Methods:The patients with malignant tumors who were using EGFRIs were divided into treatment group and control group according to whether suffering from pruritus.The concentrations of inflammation cytokines in peripheral venous blood were analyzed by human antibody array.Results:The differential inflammatory cytokines were(granulocyte-macrophage colony-stimulating factor, GM-CSF),IL-16 and(recombinant human macrophage colony stimulating factor, MCSF).Pathway enrichment showed the response to macrophage colony-stimulating factor was related to EGFRIs-induced pruritus.Conclusion:The macrophage inflammatory response induced by GM-CSF may be involved in the occurrence of EGFRIs-related pruritus.
