槲皮素通过抑制溶质载体家族7成员11表达诱导前列腺癌细胞铁死亡的临床研究 被引量:2

摘要 目的 探讨槲皮素对溶质载体家族7成员11(SLC7A11)的调节作用,分析其对前列腺癌(PC)细胞铁死亡的影响。方法 将对数期LNCaP细胞株随机分为对照组、槲皮素低剂量组、槲皮素高剂量组、槲皮素高剂量+SLC7A11阴性转染组、槲皮素高剂量+SLC7A11过表达组。细胞计数试剂盒-8(CCK-8)法检测各组LNCaP细胞的增殖情况;流式细胞仪检测各组LNCaP细胞凋亡情况;免疫印迹法检测SLC7A11、谷胱甘肽过氧化物酶4(GPX4)、p53相关蛋白表达情况;试剂盒法检测丙二醛(MDA)含量情况。结果 与对照组比较,槲皮素低剂量组、槲皮素高剂量组LNCaP细胞增殖抑制率、凋亡率、MDA、p53蛋白升高(P<0.05),SLC7A11、GPX4相关蛋白表达水平降低(P<0.05),且槲皮素低剂量组、槲皮素高剂量组间上述各项指标比较,差异具有统计学意义(P<0.05);与槲皮素高剂量组、槲皮素高剂量+SLC7A11阴性转染组比较,槲皮素高剂量+SLC7A11过表达组LNCaP细胞增殖抑制率、凋亡率、MDA、p53降低(P<0.05),SLC7A11、GPX4相关蛋白表达水平升高(P<0.05)。结论 槲皮素可能通过抑制SLC7A11表达诱导PC细胞铁死亡。 Objective To investigate the regulatory effect of quercetin on solute carrier family 7 member 11(SLC7A11),and its effect on ferroptosis of prostate cancer cells.Methods The logarithmic phase LNCaP cell line was randomly divided into control group,low-dose quercetin group,high-dose quercetin group,high-dose quercetin+SLC7A11 negative transfection group,and high-dose quercetin+SLC7A11 overexpression group.Cell counting kit-8(CCK-8) method was used to detect the proliferation of LNCaP cells in each group;flow cytometry was used to detect the apoptosis of LNCaP cells in each group;Western blotting was used to detect the expression of SLC7A11,glutathione peroxidase 4(GPX4),and p53 gene(p53) related proteins;kit method was used to detect the content of malondialdehyde(MDA).Results Compared with the control group,the LNCaP cell proliferation inhibition rate,apoptosis rate,MDA and p53 proteins were increased in the low-dose quercetin group and high-dose quercetin group(P<0.05),and the expression levels of SLC7A11 and GPX4 related proteins were reduced(P<0.05),and the above indicators were compared between the low-dose quercetin group and high-dose quercetin group and the difference was statistically significant(P<0.05).Compared with the high-dose quercetin group and the high-dose quercetin+SLC7A11 negative transfection group,the LNCaP cell proliferation inhibition rate,apoptosis rate,MDA,and p53 in the high-dose quercetin+SLC7A11 overexpression group decreased(P<0.05),the expression levels of SLC7A11 and GPX4 related proteins were increased(P<0.05).Conclusions Quercetin may induce the ferroptosis of prostate cancer cells by inhibiting the expression of SLC7A11.
