宫颈癌组织中人乳头瘤病毒16型E6、人乳头瘤病毒16型E7蛋白表达及DNA病毒载量与调节性T细胞浸润的相关性分析 被引量:2

摘要 目的 探究宫颈癌组织中人乳头瘤病毒16型(HPV16)E6、HPV16 E7蛋白表达及DNA病毒载量与调节性T(Treg)细胞浸润的相关性及临床意义。方法 选取成都医学院第一附属医院2018年1月至2020年1月84例行手术治疗的宫颈癌患者的癌组织作为宫颈癌组,选取同一医院同期手术治疗的42例宫颈上皮内瘤变(CIN)患者的宫颈组织作为CIN组,另选取42例因子宫肌瘤行全子宫切除术的患者的正常宫颈组织作为正常组。对比分析三组组织中HPV16 E6、HPV16 E7蛋白表达及DNA病毒载量、Treg细胞浸润的情况。结果 宫颈癌组HPV16 E6、HPV16 E7蛋白阳性表达率及DNA病毒载量、Treg细胞占CD4~+T细胞比例均高于CIN组、正常组,CIN组上述指标均高于正常组(P<0.05);宫颈癌组织中HPV16 E6、HPV16 E7蛋白阳性表达率及DNA病毒载量与Treg细胞占CD4~+T细胞比例呈正相关(P<0.05);HPV16 E6、HPV16 E7蛋白阳性表达率及DNA病毒载量、Treg细胞占CD4~+T细胞比例与患者临床分期呈正相关,与组织学分级呈负相关(P<0.05)。结论 宫颈癌组织中HPV16 E6、HPV16 E7蛋白表达及DNA病毒载量与Treg细胞浸润具有正相关关系,且均与患者临床分期、组织学分级显著相关,可能对宫颈癌进展有联合预测价值。 Objective To explore the correlation and clinical significance of human papilloma virus type 16(HPV16) E6 and HPV16 E7 protein expression,DNA viral load and regulatory T(Treg) cell infiltration in cervical cancer tissues.Methods The cancer tissues of 84 patients with cervical cancer who underwent surgical treatment in the First Affiliated Hospital of Chengdu Medical College from January 2018 to January 2020 were selected as the cervical cancer group,and 42 patients with cervical intraepithelial neoplasia(CIN) who were surgically treated in the same hospital during the same period were selected as the CIN group,and 42 patients who underwent total hysterectomy due to uterine fibroids were included in the normal group.The expression of HPV16 E6 and HPV16 E7 protein,DNA viral load and Treg cell infiltration in the three groups of tissues were compared and analyzed.Results The positive expression rate of HPV16 E6 and HPV16 E7 protein,DNA viral load,and the proportion of Treg cells in CD4^(+) T cells in cervical cancer group were higher than those in CIN group and normal group,and whose in the CIN group was higher than those in normal group(P<0.05).The positive expression rates of HPV16 E6 and HPV16 E7 proteins,and DNA viral load were positively correlated with the proportion of Treg cells in CD4^(+) T cells;the positive expression rates of HPV16 E6 and HPV16 E7 proteins,DNA viral loads,and the proportion of Treg cells in CD4^(+) T cells was positively correlated with clinical stage and negatively correlated with histological grade(P<0.05).Conclusions HPV16 E6,HPV16 E7 protein expression,DNA viral load in cervical cancer tissues are positively correlated with the Treg cell infiltration,and are all significantly correlated with clinical staging and histological grading,and may have a joint predictive value for cervical cancer progression.
