稽留流产患者绒毛组织中同源框基因A10 mRNA、凋亡相关调控基因p53 mRNA的表达及其临床意义 被引量:1

摘要 目的探究同源框基因A10(HOXA10)mRNA、凋亡相关调控基因p53 mRNA在稽留流产(MA)患者绒毛组织中的表达水平及意义。方法将2019年6月至2021年9月监利市人民医院收治的75例MA患者纳入MA组,并将同期行人工流产的75例正常早孕者纳入对照组。采用实时荧光定量聚合酶链反应(qRT-PCR)法检测两组绒毛组织中HOXA10 mRNA、p53 mRNA的表达水平;采用Pearson法分析MA患者绒毛组织中HOXA10 mRNA与p53 mRNA的相关性;采用受试者工作特征(ROC)曲线评价绒毛组织中HOXA10 mRNA与p53 mRNA表达水平对MA的诊断价值。结果与对照组相比,MA组患者绒毛组织中HOXA10 mRNA表达水平明显降低,p53 mRNA表达水平明显升高(P<0.05);MA患者绒毛组织中HOXA10 mRNA与p53 mRNA呈负相关(r=-0.453,P<0.05);绒毛组织中HOXA10 mRNA、p53 mRNA诊断MA的曲线下面积(AUC)分别为0.875、0.887,截断值分别为0.77、1.41,灵敏度均为88.0%,特异度分别为80.0%、74.7%;两者联合诊断MA的AUC为0.962,其灵敏度、特异度分别为86.7%、92.0%。结论MA患者绒毛组织中HOXA10 mRNA表达水平降低,p53 mRNA表达水平升高,两者呈负相关,且HOXA10 mRNA、p53 mRNA联合可有效提高对MA的诊断价值。 Objective To investigate the expression levels and significance of homeobox gene A10(HOXA10) mRNA and apoptosis related regulatory gene p53 mRNA in villous tissue of patients with missed abortion(MA).Methods A total of 75 cases of MA patients in Jianli People′s Hospital from June 2019 to September 2021 were selected for the study(MA group),and 75 cases of normal early pregnancy women who underwent artificial abortion in the same period were included as the control group.The expression levels of HOXA10 mRNA and p53 mRNA in villous tissue were detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR);Pearson method was used to analyze the correlation between the expression level of HOXA10 mRNA and p53 mRNA;receiver operating characteristic(ROC) curve was used to evaluate the diagnostic value of HOXA10 mRNA and p53 mRNA expression levels in villous tissue for MA.Results Compared with the control group,the expression level of HOXA10 mRNA in villous tissue of MA group was significantly decreased,and the expression level of p53 mRNA was significantly increased(P<0.05);there was a negative correlation between the expression of HOXA10 mRNA and p53 mRNA in MA patients(r=-0.453,P<0.05);the area under the curve(AUC) of HOXA10 mRNA and p53 mRNA in villous tissue in the diagnosis of MA was 0.875 and 0.887 respectively,and the cut-off value was 0.77 and 1.41 respectively,the corresponding sensitivity was 88.0%,and the specificity was 80.0% and 74.7%,respectively;the AUC of the combined diagnosis was 0.962,the sensitivity and specificity were 86.7% and 92.0% respectively.Conclusions The expression level of HOXA10 mRNA in villous tissue of MA patients is lower,the expression of p53 mRNA is higher,and there is a negative correlation between them.The combination of HOXA10 mRNA and p53 mRNA can effectively improve the diagnostic value of MA.
