有氧运动对糖尿病大鼠骨骼肌AMPK和GLUT4蛋白表达的影响

The Effects of Aerobic Exercise on AMPK and GLUT4 Protein Expression in Skeletal Muscle of Diabetic Rats

摘要 目的:探讨有氧运动对糖尿病大鼠骨骼肌AMPK和GLUT4蛋白表达的影响,分析运动改善糖尿病大鼠胰岛素抵抗的机制。方法:雄性SD大鼠40只,除正常对照组(NC),其余大鼠经高脂饲养和注射链尿佐菌素建立糖尿病大鼠模型,再随机分为糖尿病安静组(DM)和糖尿病运动组(DME)。DME组大鼠进行6次/周、1次/天、60min/次的无负重游泳运动干预8周。采集大鼠血液、股四头肌等组织,并检测FBG、FINS、血脂(TG、TC、LDL-C、HDL-C)、骨骼肌AMPK和GLUT4蛋白表达。结果:与NC组相比,DM组大鼠FBG、FINS、TG、TC均显著升高(P <0.01),胰岛素敏感性(ISI)、AMPK和GLUT4蛋白表达均显著降低(P <0.05);与DM组相比,DME组大鼠FBG、FINS、TG、TC均显著降低(P <0.05),ISI、AMPK和GLUT4蛋白表达均显著升高(P <0.05)。结论:8周游泳运动能够较好地控制糖尿病大鼠血糖、血脂水平,提高胰岛素敏感性;有氧运动能够增加大鼠骨骼肌AMPK和GLUT4蛋白表达,改善糖尿病大鼠胰岛素抵抗。 OBJECTIVE:The paper present a study of the effects of aerobic exercise on the expression of AMPK and GLUT4 protein in skeletal muscle of diabetic rats,and an analysis of the mechanism of exercise improving insulin resistance in diabetic rats.METHODS:40 male SD rats,except normal control(NC)group,were fed with high fat and injected with streptozotocin to establish diabetic rat model,and then randomly divided into diabetes mellitus(DM)quiet group and diabetes mellitus exercise(DME)group.Rats in DME group were subjected to weightless swimming 6 times/week,once a day,60 min/time for 8 weeks.Blood,quadriceps femoris and other tissues were collected,and protein expressions of FBG,FINS,blood lipids(TG,TC,LDL-C,HDL-C),AMPK and GLUT4 in skeletal muscle were detected.RESULTS:Compared with NC group,FBG,FINS,TG and TC in DM group were significantly increased(P<0.01),and the protein expressions of insulin sensitivity index(ISI),AMPK and GLUT4 were significantly decreased(P<0.05).Compared with DM group,FBG,FINS,TG and TC in DME group were significantly decreased(P<0.05),and the protein expressions of ISI,AMPK and GLUT4 were significantly increased(P<0.05).CONCLUSION:Swimming exercise for 8 weeks can effectively control blood glucose and lipid levels and increase insulin sensitivity of diabetic rats.Aerobic exercise can increase the protein expression of AMPK and GLUT4 in skeletal muscle and improve insulin resistance in diabetic rats.
