升阳益胃汤对慢性胃炎小鼠胃组织病理变化及血清IL-2、IL-6的影响

Effects of Shengyang Yiwei Decoction(升阳益胃汤)on Gastric Mucosal Pathological Changes and Serum IL-2,IL-6 Levels in Chronic Gastritis Mice

目的:探讨升阳益胃汤对慢性胃炎小鼠胃组织病理变化及血清炎症因子的影响,初步阐释升阳益胃汤治疗慢性胃炎的作用机制;为该方治疗慢性胃炎提供实验依据。方法:将40只雄性ICR小鼠随机分为正常组、模型组、升阳益胃汤组、西药组,每组10只。以2%水杨酸钠2 mL每天灌胃结合饥饿饱餐因素共8周制备慢性胃炎小鼠模型。造模结束后各组小鼠给予相应药物灌胃30 d,取胃标本采用HE染色,光镜观察胃组织病理变化,ELISA检测各组小鼠血清IL-2、IL-6水平。结果:正常组小鼠胃组织未见炎性病理改变;模型组胃部可见胃黏膜炎症表现;中药组炎症程度明显减轻。ELISA检测结果显示模型组小鼠血清IL-2表达水平低于正常组,中药组IL-2表达水平高于模型组但低于正常组(P<0.05);模型组小鼠血清IL-6表达水平高于正常组,中药组IL-6表达水平低于模型组但高于正常组(P<0.05)。结论:升阳益胃汤能减轻慢性胃炎小鼠的胃黏膜病理损伤;调节血清IL-2和IL-6等炎症因子分泌是其作用机制之一。

Objective:To explore the effect of Shengyang Yiwei Decoction on gastric histopathological changes and inflammatory factors in mice with chronic gastritis,and to preliminarily explain the mechanism of Shengyang Yiwei Decoction in treating mice with chronic gastritis;to provide experimental basis for the treatment of chronic gastritis.Methods:40 male ICR mice were randomly divided into normal group,model group,Shengyang Yiwei Decoction group and Western medicine group,with 10 mice in each group.The mouse model of chronic gastritis was prepared by daily gavage of 2%sodium salicylate for 8 weeks.After modeling,each group was given the corresponding medicine for 3 days,the pathological changes of gastric tissue were evaluated by HE staining and observed by microscope,and the changes of serum cytokines IL-2 and IL-6 were detected by ELISA.Results:There were no inflammatory pathological changes in gastric mucosal in normal group;gastric mucosal inflammation was observed in the model group;the degree of inflammation was significantly reduced in Shengyang Yiwei Decoction group.The results of ELISA showed that the level of serum IL-2 in normal group was higher than that in model group,and that in Shengyang Yiwei Decoction group was higher than that in model group but lower than that in normal group(P<0.05).The level of serum IL-6 in model group was higher than that in normal group,and that in Shengyang Yiwei Decoction group was lower than that in model group but higher than that in normal group(P<0.05).Conclusion:Shengyang Yiwei Decoction can reduce the pathological damage of gastric mucosa in mice with chronic gastritis;that may be related to regulating inflammatory factors such as IL-2 and IL-6.
