摘要
目的:探讨甘麦龙骨汤对慢性睡眠剥夺大鼠神经递质及激素水平的影响。方法:44只雄性SD大鼠随机分入正常对照组,正常中药组,模型对照组,及模型+中药组,每组11只;通过改良多平台水环境法建立慢性睡眠剥夺模型。正常对照组及模型组予生理盐水灌胃;中药组及模型+中药组予甘麦龙骨汤灌胃(18 g/kg,1次/天,连续10 d),干预结束后采用Y迷宫实验进行动物行为学评价。行为学实验后立即麻醉大鼠并抽取脑脊液及血清,ELISA对相关递质及激素水平进行测定。结果:①动物行为学实验结果:与正常对照组比较,模型组错误次数明显增多,主动回避率明显减少,差异有统计学意义(P<0.05);与模型组相比,模型+中药组错误次数明显减少,主动回避率增多,差异有统计学意义(P<0.05)。②脑脊液及血清相关递质及激素水平检测结果:与正常对照组比较,模型对照组大鼠脑脊液及血清前列腺素D2(PGD2)水平以及血清五羟色胺(5-HT)、促甲状腺激素(TSH)、去甲肾上腺素(NE)、皮质醇(CORT)水平显著升(P<0.05);与模型组相比,模型+中药组大鼠脑脊液及血清PGD2及血清5-HT、TSH、NE、CORT水平显著降低(P<0.05)。模型+中药组大鼠与正常对照组、正常中药组大鼠各指标检测结果差异均不显著(P<0.05)。结论:甘麦龙骨汤的促眠机制可能与其下调脑脊液及外周血清PGD2及血清5-HT、TSH、NE、CORT水平有关。
Objective:To investigate the effects of Ganmai Longgu Decoction in rats with chronic sleep deprivation based on the neurotransmitter and hormone levels.Methods:Forty-four male SD rats were randomly divided into control group,Chinese medicine group,model group and model+Chinese medicine group,with each 11 rats.Models with chronic sleep deprivation were established via a modified multi-platform water environment method.The control group and model group were intervened by intragastrical administration of normal saline,while another two groups were intervened by intragastrical administration of Ganmai Longgu Decoction(18 g/kg,1/d,for 10 consecutive days).Animal behavior evaluation was performed follows interventions.Immediately after the behavioral test,the rats were anesthetized to extract cerebrospinal fluid(CSF)and serum,and a variety of neurotransmitters and hormones levels were measured via ELISA.Results:①The results of animal behavior experiments showed that compared with the control group,the number of errors in the model group was significantly increased,and the rate of active avoidance was significantly reduced,the difference was statistically significant(P<0.05);Compared with the model group,the number of errors in the model+Chinese medicine group was significantly reduced,The rate of active avoidance increased,and the difference was statistically significant(P<0.05).②The results of neurotransmitters and hormones in CSF and serum showed that compared with the control group,both CSF and serum levels of Prostaglandin D2(PGD2)as well as the serum levels of 5-hydroxytryptamine(5-HT),thyroid stimulating hormone(TSH),norepinephrine(NE)and cortisol(CORT)were much higher in the model group(P<0.05).Compared with the model group,both CSF and serum levels of PGD2 as well as the serum levels of 5-HT,TSH,NE and CORT were much lower in the model+Chinese medicine group(P<0.05).No significant difference in each index was found among the control,Chinese medicine,and model+Chinese medicine group.Conclusion:The sleep-promoting effects of Ganmai Longgu Decoction may be associated with its down-regulation of serum 5-HT,TSH,NE,CORT levels as well as PGD2 levels in both CSF and serum.
