甘草甜素对缺氧复氧损伤心肌细胞的保护作用及其lnc-SNHG5表达的影响 被引量:1

摘要 目的:研究甘草甜素对心肌缺氧复氧损伤的保护作用及其与小核仁RNA宿主基因5(lnc-SNHG5)表达的相关性。方法:H9C2心肌细胞分为4组,正常对照组(Control)、模型组(Hypoxia Reoxygenation,HR)、转染组(Transfection group,TG)、甘草甜素组(Glycyrrhizin group,GP),其中模型组、转染组、甘草甜素组均构建心肌细胞缺氧复氧模型,转染组预先腺病毒转染沉默lnc-SNHG5表达,甘草甜素组预先给药,浓度为40μmol/L,CCK8检测细胞存活率;流式细胞仪检测细胞凋亡,qPCR检测lnc-SNHG5表达。结果:与模型组比较,转染组与甘草甜素组细胞存活率均提高(均P<0.05),细胞凋亡率均下降(均P<0.05);与正常对照组比较,模型组lnc-SNHG5表达显著升高(P<0.05),甘草甜素组相较于模型组,lnc-SNHG5表达显著降低(P<0.05)。结论:甘草甜素可能通过抑制lnc-SNHG5的表达发挥对心肌细胞缺氧复氧损伤的保护作用。 Objective:To research the protective effect of Glycyrrhizin on myocardial hypoxia and reoxygenation injury and its correlation with lnc-SNHG5 expression.Methods:H9C2 cardiomyocytes were cultured and divided into 4 groups:Control group,Model group(Hypoxia Reoxygenation,HR),Transfection group(TG),Glycyrrhizin group(GP),The HR group,TG group and GP group were all constructed myocardial hypoxia and reoxygenation models.The TG group was pre-transfected with adenovirus to silence lnc-SNHG5 expression,and the Glycyrtin group was pre-administered with the concentration of 40μmol/L,and the cell survival rate was analyzed by CCK8.Cell apoptosis was detected by flow cytometry and lnc-SNHG5 expression was detected by qPCR.Results:Compared with HR group,the survival rate of TG group and GP group was increased,and the apoptosis rate was decreased(all P<0.05).Compared with Control group,the expression of lnc-SNHG5 in HR group was significantly increased(P<0.05),and the expression of lnc-SNHG5 in GP group was significantly decreased compared with HR group(P<0.05).Conclusion:Glycyrrhizin may protect cardiomyocytes from hypoxia and reoxygenation injury by inhibiting lnc-SNHG5 expression.
