长期低浓度吸入七氟醚对成年小鼠肝、肾功能的影响

摘要 目的探讨长期低浓度七氟醚暴露对成年小鼠肝、肾功能的影响,为麻醉医生及手术室工作人员职业暴露安全性提供参考。方法8周龄KM小鼠24只,随机均分为对照组、0.1%七氟醚组和0.3%七氟醚组,分别暴露于空气、0.1%七氟醚和0.3%七氟醚环境中,1 h/d,5 d/周,共12周。计算肝肾系数,检测血清中肝、肾功能生化指标,检测胰岛素样生长因子-1(IGF-1)水平,检测肝脏组织中凋亡蛋白的表达,观察肝肾组织细胞病理学改变。结果3组小鼠体质量、肝肾系数、肝肾功能及凋亡蛋白比值差异均无统计学意义(P>0.05);IGF-1表达水平随着吸入七氟醚浓度的增加而下降(P<0.05);肝脏病理学显示随吸入七氟醚浓度的增加,病理损伤加重;肾脏病理学显示3组小鼠间差异无统计学意义(P>0.05)。结论长期暴露于低浓度七氟醚环境会导致成年小鼠肝脏产生一定的损伤而未对肾脏产生影响。 Objective To investigate effects of long time indraft of sevoflurane at low concentration on liver and renal function in adult rat,in order to provide reference for the safety with occupational exposure and occupational protection of anesthesiologists and operating room staff.Methods Adult KM rat aged 8 weeks were randomly divided into a control group,a 0.1%sevoflurane group and a 0.3%sevoflurane group.Rats in the three groups were exposed to air,0.1%sevoflurane and 0.3%sevoflurane for 1 h/d and 5 days/week for 12 consecutive weeks,respectively,to calculate the liver and renal coefficient of mice,test their liver and renal function,insulin-like growth factor-1(IGF-1)level in serum and the expression of apoptin in the liver tissue,and observe the paohological changes of issue cells.Results There were no significant differences in body weight,liver and renal coefficient,function and apoptotic protein ratio of mice(P>0.05).The expression level of IGF-1 decreased with the increase of inhalation concentration.Conclusion Long time indraft of sevoflurane at low concentration can damage liver functions in adult mice to a certain extent but with no effect on kidneys.
