大黄素阻断SMAD2/3信号通路抑制人肺动脉平滑肌细胞增殖和迁移

Emodin inhibits the proliferation and migration of human pulmonary artery smooth muscle cells by blocking SMAD2/3 signaling pathway

摘要 目的探讨大黄素对转化生长因子β1(TGF-β1)诱导的人肺动脉平滑肌细胞(HPASMC)增殖、迁移的影响。方法体外培养HPASMC,取对数生长期的细胞,分为对照组、TGF-β1处理组、TGF-β1联合大黄素处理组。采用CCK-8法检测HPASMC活性,Transwell^(TM)小室实验和划痕实验检测HPASMC迁移能力;免疫荧光实验检测骨桥蛋白(OPN)、增殖细胞核抗原(PCNA)的表达,Western blot法检测HPASMC的OPN、PCNA的蛋白水平及SMAD家族成员2(SMAD2)和SMAD3的磷酸化水平。结果与对照组相比,TGF-β1处理组的OPN、PCNA蛋白表达增加,HPASMC增殖和迁移能力增强,SMAD2、SMAD3的磷酸化增强。与TGF-β1组相比,TGF-β1联合大黄素处理组HPASMC的增殖和迁移能力降低,OPN、PCNA蛋白表达减少,SMAD2、SMAD3蛋白磷酸化明显降低。结论大黄素可抑制HPASMC中TGF-β1上调OPN、PCNA蛋白的表达,抑制TGF-β1诱导的PASMC增殖和迁移,可能与抑制SMAD2/3信号通路有关。 Objective To investigate the effect of emodin on the proliferation and migration of human pulmonary artery smooth muscle cells(HPASMCs)induced by transforming growth factorβ1(TGF-β1).Methods HPASMCs were cultured in vitro,and HPASMCs in logarithmic growth phase were divided into control group,TGF-β1 group,and TGF-β1 combined with emodin group.The activity of HPASMCs was detected by CCK-8 assay,the migration ability of HPASMCs was detected by Transwell^(TM) chamber assay and scratch assay,and the expressions of osteopontin(OPN)and proliferating cell nuclear antigen(PCNA)were detected by immunofluorescence assay.The protein levels of OPN and PCNA and the phosphorylation of SMAD family member 2(SMAD2)and SMAD family member 2(SMAD3)in HPASMCs were detected by Western blot.Results Compared with those in the control group,in TGF-β1 group,the protein expressions of OPN andPCNA,the proliferation and migration of HPASMCs,and the phosphorylation of SMAD2 and SMAD3 were increased.Compared with those in the TGF-β1 group,in the TGF-β1 combined with emodin group,the proliferation and migration of HPASMCs,the expressions of OPN and PCNA,and the phosphorylation of SMAD2 and SMAD3 were decreased.Conclusion Emodin inhibits the up-regulation of OPN and PCNA and the proliferation and migration of PASMCs induced by TGF-β1,which may be related to the blocking of SMAD2/3 signaling pathway.
