摘要
为探索木麻黄的小枝茎段在离体条件下培养的可行性、再生方式及其形态特征的变化,对木麻黄离体小枝茎段器官的培养及其再生植株的发生与构建进行了研究。结果表明:半木质化的小枝适宜培养,为较好的外植体材料。适宜的继代培养基为改良MS+6BA0.25+NAA0.1+糖30g·L^(−1)+卡拉胶5.5g·L^(−1),不定根诱导培养基为1/2MS+NAA0.5+糖20g·L^(−1)。
In order to explore the feasibility of in vitro culture of Casuarinaequisetifolia,the regeneration mode,morphological characteristics,shoot proliferation and development of in vitro propagated plants were studied.The results showed that the semi-lignified twigs were suitable for culture and lead to better quality explants.The optimal subculture medium was found to be modified MS+6BA0.25+NAA0.1+sugar 30 g·L^(−1)+carrageenan 5.5 g·L^(−1),and the best adventitious root induction medium was found to be 1/2MS+NAA0.5+sugar 20 g·L^(−1).
