摘要 目的 探究2型细胞因子白细胞介素4(interleukin-4,IL-4)和IL-13对慢性鼻窦炎伴鼻息肉(chronic rhinosinusitis with nasal polyps,CRSwNP)中鼻黏膜上皮细胞自噬的影响。方法 收集16例CRSwNP息肉组织(其中嗜酸细胞型和非嗜酸细胞型鼻息肉组织各8例)和8例对照黏膜组织。Western blot检测自噬标志蛋白——人微管相关蛋白1A/1B轻链3B(LC3B)的表达,免疫组化染色检测LC3B的表达和分布,免疫荧光染色实验检测LC3B的点状分布。结果 与对照黏膜组织相比,LC3BⅡ的表达在CRSwNP的息肉组织中增加,并且LC3B在嗜酸细胞型鼻息肉上皮层分布最多。IL-4和IL-13可诱导鼻黏膜上皮细胞中LC3BⅡ的表达和促进LC3B的点状分布。结论 与对照黏膜和非嗜酸细胞型鼻息肉组织相比,嗜酸细胞型鼻息肉组织上皮细胞中自噬体数量显著增加。IL-4和IL-13可有效诱导鼻黏膜上皮细胞自噬体增加。本研究为寻找以自噬为靶点的CRSwNP治疗新方法提供理论依据。 OBJECTIVE To explore the effect of type 2 cytokines IL-4 and IL-13 on autophagy of nasal epithelial cells in chronic rhinosinusitis with nasal polyps(CRSwNP).METHODSWe collected nasal polyp tissue from 16 patients with CRSwNP(8 eosinophilic nasal polyp and 8 non-eosinophilic nasal polyp) and normal mucosa tissue from 8 healthy control.Western blot was used to detect the expression of autophagy marker protein LC3B.Immunohistochemical staining was used to detect the expression and distribution of LC3B.Immunofluorescence staining was used to detect the LC3B puncta.RESULTS The expression of LC3BⅡ was increased in polyp tissue of CRSwNP compared with control mucosal tissue,and LC3B was most distributed in the nasal epithelial cells of eosinophilic nasal polyps.IL-4 and IL-13 could induce the expression of LC3BⅡ and LC3B puncta in human nasal epithelial cells(HNEPCs).CONCLUSION The number of autophagosomes was significantly increased in the epithelial cells of eosinophilic nasal polyp tissues compared with control mucosa and non-eosinophilic nasal polyp tissues.IL-4 and IL-13 could effectively increase the number of autophagosomes in HNEPCs.Our study will provide a theoretical basis for discovering a novel autophagy-targeting therapeutic method for CRSwNP treatment.
