头花蓼提取物对左氧氟沙星血浆蛋白结合率的影响 被引量:1

摘要 目的考察头花蓼提取物对左氧氟沙星血浆蛋白结合率的影响。方法采用超高效液相-串联质谱法(UPLC-MS/MS)测定左氧氟沙星含量,色谱柱为Waters BEH C_(18)(2.1 mm×100 mm,1.7μm)柱,流动相为0.1%甲酸乙腈-0.1%甲酸水,柱温为45℃,流速为0.30 mL·min^(-1),进样体积为1μL。以中药灌胃预处理大鼠后,分离血浆与左氧氟沙星作用,通过平衡透析法考察左氧氟沙星在3μg·mL^(-1)、6μg·mL^(-1)两种浓度下,头花蓼提取物对左氧氟沙星血浆蛋白结合率的影响。结果左氧氟沙星在血浆及透析外液浓度范围内线性关系良好,其精密度、准确度、稳定性、提取回收率及基质效应均符合生物样品分析测定要求。左氧氟沙星单独与大鼠血浆作用,血浆蛋白结合率分别为(22.53±3.16)%、(20.03±8.00)%;大鼠灌胃头花蓼提取物预处理后,分离的血浆与左氧氟沙星蛋白结合率显著增加,分别增加至(43.95±4.14)%、(35.27±6.05)%(P<0.05)。结论头花蓼提取物能够使左氧氟沙星的血浆蛋白结合率升高。 Objective To determine the effect of Polygonum capitatum extract on the plasma protein binding rate of levofloxacin.Methods Ultra-performance liquid chromatography-tandem mass spectroscopy(UPLC-MS/MS)was used to determine the content of levofloxacin.A Waters BEH C_(18)(2.1 mm×100 mm,1.7μm)column was used.The mobile phase was 0.1% formic acid acetonitrile-0.1% formic acid water,the column temperature was 45℃,the flow rate was 0.30 mL·min^(-1),and the injection volume was 1μL.After pretreatment of rats by gavage with traditional Chinese medicine,the separated plasma interacted with levofloxacin.The effect of Polygonum capitatum extract on the protein binding rate of levofloxacin(3μg·mL^(-1) and 6μg·mL^(-1))was investigated by equilibrium dialysis method.Results Levofloxacin showed a good linearity in the concentration of the plasma and dialysis fluid.The precision,accuracy,stability,extraction recovery and matrix effect all met the requirements of biological sample determination.Levofloxacin alone interacted with the rat plasma,and the plasma protein binding rates were(22.53±3.16)% and(20.03±8.00)%,respectively.After the pretreatment with Polygonum capitatum extract,the protein binding rate of separated plasma and levofloxacin increased significantly to(43.95±4.14)% and(35.27±6.05)%,respectively(P<0.05).Conclusion Polygonum capitatum extract can significantly increase the plasma protein binding rate of levofloxacin.
