摘要
目的:观察枳术通便汤对便秘小鼠排便功能及结肠病理变化的影响。方法:将60只美国癌症研究所(Institute of Cancer Research,ICR)雄性小鼠随机分为正常组、模型组、阳性药莫沙必利组、枳术通便汤高、中、低剂量组6组。采用复方地芬诺酯混悬液对除正常组以外的5组小鼠灌胃14 d,建立便秘模型。分别以枳术通便汤、莫沙必利对枳术通便汤高、中、低剂量组和莫沙必利组给药治疗14 d。分别于造模前后、治疗前后检测首次排便时间及24 h内粪便粒数及粪便总质量、粪便含水率,检测小肠推进率,运用酶联免疫吸附试验法检测小鼠结肠组织血管活性肠肽(Vasoactive Intestinal Peptide,VIP)、P物质(Substance P,SP)表达的影响。苏木素-伊红染色,光学显微镜下观察实验小鼠结肠病理变化。结果:①与模型组比较,枳术通便汤可以显著缩短首粒粪便时间(P<0.05),同时可以增加小鼠粪便总质量及粪便含水率(P<0.05)。②酶联免疫吸附试验法检测结果显示,与正常组比较,模型组抑制性神经递质VIP含量显著升高,SP含量显著降低(P<0.05)。与模型组比较,枳术通便汤高、中剂量组以及阳性药莫沙必利组VIP含量显著降低,SP含量显著升高(P<0.05),但枳术通便汤低剂量组VIP、SP含量的差异无统计学意义。③枳术通便汤可以使便秘小鼠结肠组织黏膜炎性渗出显著减轻。结论:枳术通便汤可能是通过上调结肠组织中SP的含量,下调结肠组织中VIP的表达来改善实验小鼠的便秘症状,同时还具有保护便秘小鼠结肠黏膜损伤的作用。
Objective:To observe the effect of the Zhizhu Tongbian decoction(枳术通便汤)on defecation function and colon pathological changes in constipated mice.Methods:Sixty ICR’s mice were randomly divided into 6 groups:the blank group,the model group,the mosapride group,and the Zhizhu Tongbian decoction high,medium and low dose groups.The constipation model was established by gavage in 5 groups except the blank group with compound diphenoxylate suspension for 14 days.The Zhizhu Tongbian decoction and mosapride were administered to the Zhizhu Tongbian decoction high,medium and low dose groups and the mosapride group for 14 days,respectively.Before and after modeling and before and after treatment,the time of first defecation and the number of fecal particles,total fecal mass,and fecal moisture content within 24 hours were detected,respectively.The propulsion rate of the small intestine was detected,and the expression of VIP and SP in the colon tissue of mice was detected by ELISA.HE staining was used to observe the pathological changes of the colon of experimental mice under a light microscope.Results:①Compared with the model group,the Zhizhu Tongbian decoction significantly shortened the time to the first feces(P<0.05),and at the same time increased the total mass and water content of feces in mice(P<0.05).②ELISA test results:compared with the normal group,the content of inhibitory neurotransmitter VIP in the model group was significantly increased,and the content of SP was significantly decreased(P<0.05).Compared with the model group,the VIP content in the Zhizhu Tongbian decoction high and middle dose groups and the mosapride group were significantly decreased,while the SP contents were increased significantly(P<0.05),but there was no significant difference in the contents of VIP and SP in the Zhizhu Tongbian decoction low-dose group.③The Zhizhu Tongbian decoction can significantly reduce the inflammatory exudation of colonic tissue in mice with constipation.Conclusion:The Zhizhu Tongbian decoction may ameliorate the symptoms of constipation in experimental mice by up-regulating the content of SP in colon tissue and down-regulating the expression of VIP in colon tissue,and also has the effect of protecting colonic mucosal in constipated mice.
