摘要
以野蔷薇为试验材料,采用PCR及RT-PCR的方法,研究了4CL1α1基因的基因组和CDS全长的克隆,对候选基因进行相关的生物信息学分析,以期为野蔷薇花色合成代谢通路提供参考依据。结果表明:通过半定量qRT-PCR的方法对4CL基因家族在野蔷薇不同组织中及激素诱导后的表达量进行分析。克隆得到4CL1α1基因,gDNA长度4 261 bp, CDS长度1 632 bp,编码543个氨基酸。通过生物信息学分析,初步认定其为4CL或4CL-like。通过对其中6个基因进行荧光实时定量的分析,发现4CL基因家族大多在红花和幼嫩的刺中的表达量相对较高。初步认定野蔷薇中Rm4CL基因家族在花色素合成及细胞壁增厚中发挥重要作用。
Taking the wild rose(Rosa multiflora) as the test material, the genome of the 4CL1α1 gene and the full-length CDS clone were studied by PCR and RT-PCR,in order to provide reference for the anabolic pathway of rose flower color.The results showed that the expression of the 4CL gene family in different tissues of rose and after hormone induction was analyzed by semi-quantitative qRT-PCR. The 4CL1α1 gene was cloned, with gDNA length 4 261 bp and CDS length 1 632 bp, encoding 543 amino acids. Through bioinformatics analysis, it was preliminarily determined to be 4CL or 4CL-like. Through the real-time quantification of fluorescence of 6 genes, it was found that the expression of the 4CL gene family was relatively high in safflower and young thorns. It was preliminarily determined that the Rm4CL gene family in roses plays an important role in anthocyanin synthesis and cell wall thickening.
