酒精性心肌病大鼠模型中凝集素样氧化低密度脂蛋白受体-1表达与细胞凋亡相关蛋白Bax、Cleaved caspase-3表达关系研究

Relationship between expression of LOX-1 and apoptosis-related proteins Bax,Cleaved caspase-3 in rat model of alcoholic cardiomyopathy

目的:分析酒精性心肌病(ACM)大鼠模型中凝集素样氧化低密度脂蛋白受体-1(LOX-1)的表达情况及与细胞凋亡相关蛋白Bax、Cleaved caspase-3表达量变化的相关性。方法:选用2周龄的健康雄性SD大鼠9只,随机分为三组:(1)正常对照组(Blank组);(2)酒精性心肌病组(ACM组);(3)酒精性心肌病+shRNA干预组(ACM+shRNA组),采用实时荧光定量PCR(qRT-PCR)、蛋白质印迹法(Western blot)分别在基因及蛋白水平检测LOX-1、Bax、Cleaved caspase-3的表达量并采用Pearson检验分析其相关性。结果:与Blank组相比,ACM组心肌组织中LOX-1、Bax、Cleaved caspase-3的表达量显著增加(P<0.01);与ACM组比较,ACM+shRNA组心肌组织中LOX-1、Bax、Cleaved caspase-3的表达量明显减少(均P<0.01);相关性分析提示LOX-1的表达量与Bax、Cleaved caspase-3的表达量呈正相关(r=0.323、0.370,均P<0.05)。结论:酒精性心肌病大鼠模型心肌组织中LOX-1的表达量升高,且与Bax、Cleaved caspase-3的表达量呈正相关;LOX-1可能通过上调Bax、Cleaved caspase-3的表达参与酒精诱导心肌细胞凋亡的过程。

Objective:To analyze the relationship between expression of LOX-1 and apoptosis-related proteins Bax,Cleaved caspase-3 in rat model of alcoholic cardiomyopathy.Methods:Nine healthy male SD rats at 2-week-old were selected and randomly divided into Blank group,ACM group and ACM+shRNA group(adenovirus-associated shRNA was injected intravenously on the basis of ACM group).The expression of LOX-1,Bax and Cleaved caspase-3 was detected by qRT-PCR and Western blot,and the correlation was analyzed by Pearson test.Results:Compared with the Blank group,the expressions of LOX-1,Bax and Cleaved caspase-3 were significantly increased in ACM group(all P<0.01).Compared with the ACM group,the expressions of LOX-1,Bax and Cleaved caspase-3 were significantly decreased in the ACM+shRNA group(all P<0.01).Correlation analysis suggested that LOX-1 expression was positively correlated with the Bax and Cleaved caspase-3 expression(r=0.323,0.370,all P<0.05).Conclusion:The expression level of LOX-1 was increased in alcoholic cardiomyopathy rat model,and was positively correlated with the expression level of Bax and Cleaved caspase-3.LOX-1 may be involved in alcohol-induced apoptosis of myocardial cells by up-regulating the expression of Bax and Cleaved caspase-3.
