华山松高通量全基因组SNP标记开发及其分析 被引量:2

Development and Analysis of High Flux Genome Wide SNP Markers for Armand Pine(Pinus armandii)

摘要 为开发华山松大量的稳定性高、特异性强的SNP标记,本研究以279份华山松针叶为试验材料,利用SLAF-seq技术,测序共获得Reads数据3169 Mb,SLAF标签12952676个,多态性SLAF标签为1456486个,SLAF多态性百分率是11.24%,平均测序深度为20.52 x,测序平均Q30为96.58%,平均GC含量为37.76%。且采用水稻‘日本晴’的测序数据用于评估试验建库的准确性,结果显示本试验比对效率在97%;且水稻‘日本晴’数据的酶切效率在100%,这表明本试验酶切反应也正常。最终从12952676个SLAF标签中检测到了3469074个群体SNP标记。这为华山松分子辅助育种、遗传多样性分析和遗传图谱构建等研究提供理论参考。 To develop a large number of stable and specific SNP markers for Pinus armandii,279 needles of Pinus armandii were used as experimental materials in this experiment.By using SLAF-seq technology,a total of3169 Mb Reads data,12952676 SLAF tags and 1456486 polymorphic SLAF tags were obtained.The percentage of SLAF polymorphism was 11.24%,the average sequencing depth was 20.52 x,the average Q30 was 96.58%,and the average GC content was 37.76%.In addition,the sequencing data of’Nipponbare’were used to evaluate the accuracy of the experiment.The results showed that the comparison efficiency of this experiment was 97%,and the enzyme digestion efficiency of’Nipponbare’data was 100%,which indicated that the enzyme digestion reaction was normal.Finally,3469074 SNPs were detected from 12952676 SLAF markers.This will provide a theoretical reference for molecular assisted breeding,genetic diversity analysis and genetic map construction of Pinus armandii.
