摘要
本研究以褐苞薯蓣类型的福建地方特色山药品种‘清流雪薯’试管苗为试验材料,采用固液双层的培养方式,研究外源Me JA处理对试管苗生长、试管薯形成及管外萌发的影响。结果表明:低浓度的MeJA对叶绿素含量无显著影响,在浓度大于1×10^(-6)mol/L时叶绿素含量显著下降。不同浓度的MeJA处理对试管苗株高无显著影响,但茎叶鲜重随MeJA处理浓度的增加呈先升后降的趋势,在5×10^(-8)mol/L时茎叶鲜重达最大,在1×10^(-5)mol/L时茎叶鲜重显著下降。外源Me JA处理可促进提前结薯,显著提高试管薯长度和大薯率,但对结薯率无显著影响。适宜浓度(5×10^(-7)mol/L,1×10^(-6)mol/L)的MeJA处理可以显著提高试管薯鲜重。1×10^(-7)mol/L和1×10^(-6)mol/L的Me JA处理后收获的试管薯在自然存放状态下全部萌发的时间比对照提早7 d。
The plantlets cultured in vitro of Dioscorea persimilis cv.Qingliu,a variety with local characteristics in Fujian Province,was used as experimental materials to study the effects of exogenous methyl jasmonate on the plant growth,tuberization,and tuber sprouting in vitro by using the solid-liquid double layer culture method.The results showed that low concentration of Me JA had no significant effect on the chlorophyll content of plantlets in vitro.When treated with MeJA with the concentrations higher than 1×10^(-6)mol/L,the chlorophyll content of the plantlets in vitro was significantly decreased.The concentrations of MeJA had no significant impact on the plant height,while the fresh weight of stems and leaves was initially increased and then decreased with the increased MeJA concentrations.Among the treatments,the fresh weight of stems and leaves was the heaviest under the treatment of 5×10^(-8)mol/L Me JA and the lightest under the treatment of 1×10^(-5)mol/L MeJA.The exogenous MeJA accelerated the tuberization and significantly increased the tuber length and big-sized tuber ratio in vitro,however there was no significant difference in the final tuberization rate after MeJA treatments.The optimum concentration of MeJA(5×10^(-7)mol/L and 1×10^(-6)mol/L)significantly promoted the fresh weight of microtubers.Under the normal storage conditions,the microtubers harvested from the MeJA-treated plantlets in vitro(1×10^(-7)mol/L,1×10^(-6)mol/L)sprouted seven days earlier than the control.
