The prognosis and function of HMGB3 in breast cancer:Based on bioinformatics analysis

摘要 为了探究高迁移率组盒3(HMGB3)在乳腺癌中的表达、预后作用和功能,利用TIMER数据库分析HMGB3在不同肿瘤中的表达情况和肿瘤浸润淋巴细胞对乳腺癌患者累积生存的影响,使用GEPIA和UALCAN分析HMGB3在乳腺癌和正常乳腺组织中的表达差异,用PrognoScan和Kaplan-Meier Plotter评估HMGB3在乳腺癌中的预后作用,通过Coexpedia和STRING分析HMGB3在乳腺癌中的功能.发现HMGB3 mRNA和蛋白在乳腺癌中高表达,预后分析显示HMGB3高表达预示乳腺癌患者不良预后,且HMGB3高表达在ER阳性、淋巴结阳性和TP53突变患者中的预后更差.高水平的肿瘤免疫浸润提示乳腺癌患者可以达到更好的十年累积生存.在更低的B细胞、CD8+T细胞、CD4+T细胞、自然杀伤细胞、Th1细胞浸润水平下,HMGB3高表达与乳腺癌更差的预后有关;在更高水平Treg细胞浸润下,高HMGB3表达与乳腺癌更差的预后有关.功能分析显示HMGB3与其共表达基因在乳腺癌中主要参与调控P53通路、细胞周期信号通路和药物代谢. HMGB3在乳腺癌中高表达且提示乳腺癌不良预后,可作为乳腺癌的预后指标和潜在治疗靶点. The purpose of our research is to explore the expression,prognostic effect and function of high mobility group box 3(HMGB3) in breast cancer. The TIMER database is used to analyze the expression of HMGB3 in different tumors and the effect of tumor infiltrating lymphocytes on the cumulative survival of breast cancer patients. GEPIA and UALCAN are applied to analyze the expression difference of HMGB3 between breast cancer tissues and normal breast tissues. PrognoScan and Kaplan-Meier Plotter are utilized to evaluate the prognostic value of HMGB3 in breast cancer. Coexpedia and STRING are used to analyze the function of HMGB3 in breast cancer. The mRNA and protein expression levels of HMGB3 are highly expressed in breast cancer samples. Prognostic analysis shows that high HMGB3 expression predicts poor prognosis in breast cancer patients,and has a worse prognosis in ER-positive,lymph node-positive,and TP53 mutations breast cancer patients. In breast cancer,the high level of tumor infiltrating lymphocytes suggest better 10-year cumulative survival. High expression of HMGB3 is associated with poorer prognosis in breast cancer patients with lower B cells,CD8+T cells,CD4+T cells,natural killer cells,and Th1 cells infiltration levels. High expression of HMGB3 is correlated with worse prognosis in breast cancer patients with higher Treg cell infiltration level. HMGB3 and its co-expressed genes are mainly involved in p53 signaling pathway,cell cycle signaling pathway and drug metabolism. HMGB3 is highly expressed in breast cancer and which indicates poor prognosis in breast cancer patients,and HMGB3 may be used as a prognostic indicator and potential therapeutic target for breast cancer.
