高浓度卡托普利增强阿司匹林的抗血小板作用研究

Effect of the Antiplatelet Action of Aspirin by High Concentration of Captopril

目的探究卡托普利在小鼠体内对阿司匹林抗血小板作用的影响。方法44只小鼠分别球后注射10μg·mL^(-1)阿司匹林作用1 h后,随机均分为4组,每组11只,对照组注射同体积生理盐水,低、中、高实验组分别注射不同浓度(0.033,0.33,3.3μg·mL^(-1))卡托普利。30 min后,测定并记录尾出血时间和肠系膜小动脉血栓形成时间;血小板聚集仪测定collagen诱导小鼠血小板聚集程度。结果与对照组对比,高浓度组小鼠尾出血时间显著延长(P<0.05),低浓度和中浓度组小鼠尾出血时间对比差异均无统计学意义(P>0.05);与对照组对比,高浓度组小鼠肠系统小动脉血栓形成时间较长(P<0.05),低浓度和中浓度组小鼠肠系统小动脉血栓形成时间对比差异均无统计学意义(P>0.05);对照组的中位聚集率为38%,低浓度组为32.6%,中浓度组为30%,高浓度组为21.3%,与对照组相比(中浓度组P=0.008;高浓度组P=0.002),差异具有统计学意义。结论高浓度的卡托普利能够增强阿司匹林的抗血小板作用。

OBJECTIVE To investigate the possible interactive effect of captopril on aspirin antiplatelet response in mice.METHODS Mice were intraperitoneally injected with aspirin at a dose of 10μg·mL^(-1).After one hour,Mice were divided into four groups randomly,11 in each group.The control group was re⁃injected with the same volume of normal saline,and the treatment groups were re⁃injected with captopril at a doses of 0.033,0.33,3.3μg·mL^(-1).After 30 minutes,,tail bleeding time was recorded and the occlusion time of mesenteric arteriole was measured.The platelet aggregation induced by collagen was measured by platelet aggregator.RESULTS Compared with the control group,the tail bleeding time of the mice in the high concentration group was significantly prolonged(P<0.05),and there was no significant difference in the tail bleeding time between the low concentration and medium concentration groups(P>0.05).The intestinal system small artery thrombosis time of mice in the the high concentration group was longer(P<0.05),and there was no significant difference in the intestinal system small artery thrombosis time between the low concentration and medium concentration groups(P>0.05).The median of platelet aggregation rate of the control group was 38%,and the low,middle,high concentration group was 32.6%,30%,21.3%.There had statistical significance in the middle concentration group(P=0.008)and the high concentration group(P=0.002)compared with control group.CONCLUSION High dose of captopril augments the antiplatelet action of aspirin.
