薄型子宫内膜大鼠模型microRNAs差异表达研究

摘要 目的对薄型子宫内膜大鼠模型差异microRNAs(miRNAs)进行筛选和靶基因预测,并对其进行生物信息学分析。方法采用95%乙醇宫腔灌注制备大鼠薄型子宫内膜模型,取模型组及正常对照组的大鼠子宫内膜组织进行总RNA提取、RNA质检、文库构建后,使用测序平台为illumina HiSeqTM2500进行高通量测序,以P<0.05的标准筛选差异miRNA,利用miRanda和RNAhybrid得出差异表达miRNA候选靶基因,进行基因本体(GO)、京都基因与基因组百科全书(KEGG)富集分析。结果共有52个差异miRNA,符合预测要求的有48个差异miRNA,共预测出1530个靶基因。其中表达上调的miRNA为rno-miR-375-3p、rno-miR-292-3p、rno-miR-129-1-3p、rnomiR-129-2-3p、rno-miR-291a-3p等,表达下调的miRNA为rno-miR-144-3p、rno-miR-542-3p、rno-miR144-5p等,涉及的生物过程主要有肽基-苏氨酸磷酸化修饰、细胞代谢过程调控、树突状细胞凋亡过程的调控;细胞成分主要包括细胞内细胞器、胞质、肌动蛋白细胞骨架、细胞骨架部分;分子功能主要涉及转移酶活性、小分子结合、RNA聚合酶Ⅲ转录调控区序列特异性DNA等方面。涉及的KEGG信号通路有Ras信号通路、胰岛素信号通路。结论薄型子宫内膜的发病可能与rno-miR-375-3p、rno-miR-292-3p、rno-miR-129-1-3p、rno-miR-129-2-3p、rno-miR-291a-3p、rno-miR-144-3p、rno-miR-542-3p、rno-miR-144-5p等miRNA表达相关,可能涉及的KEGG信号通路有Ras信号通路、胰岛素信号通路等。 Objective To screen and predict the target genes of differential microRNAs in a thin endometrial rat model,and to perform bioinformatics analysis on them.Methods Using 95%ethanol intrauterine perfusion to prepare a thin rat endometrial model,take the rat endometrial tissue of the model group and the normal control group for total RNA extraction,RNA quality inspection,library construction,and use the sequencing platform as illumina HiSeqTM2500 performs high-throughput sequencing,screens differential miRNAs with the standard of P<0.05,uses miRanda and RNAhybrid to obtain differentially expressed miRNA candidate target genes,and conducts GO and KEGG enrichment analysis.Results A total of 52 differential miRNAs,48 differential miRNAs that meet the prediction requirements,and a total of 1530 target genes were predicted.The miRNAs with up-regulated expression are rno-miR-375-3p,rno-miR-292-3p,rno-miR-129-1-3p,rno-miR-129-2-3p,rno-miR-291a-3p,etc.The miRNAs whose expression is down-regulated are rno-miR-144-3p,rno-miR-542-3p,rno-miR-144-5p,etc.The biological processes involved mainly involve peptidyl-threonine phosphorylation modification and cellular metabolic processes regulation,regulation of the process of dendritic cell apoptosis;cell components mainly include intracellular organelles,cytoplasm,actin cytoskeleton,and cytoskeleton parts;molecular functions mainly involve transferase activity,small molecule binding,RNA polymeraseⅢtranscription.The sequence-specific DNA and other aspects of the regulatory region.The involved KEGG signaling pathways include Ras signaling pathway and insulin signaling pathway.Conclusion The incidence of thin endometrium may be related to rno-miR-375-3p,rno-miR-292-3p,rno-miR-129-1-3p,rno-miR-129-2-3p,rno-miR-291a-3p,rno-miR-144-3p,rno-miR-542-3p,rno-miR-144-5p and other miRNAs are related to expression.The KEGG signaling pathways that may be involved include the Ras signaling pathway and the insulin signaling pathway.
