黄河鲤鱼优势腐败菌的鉴定及其腐败特性的研究 被引量:1

Identification and Spoilage Characteristics of a Dominant Bacterium from Yellow River Carp

摘要 为解决黄河鲤鱼在贮运过程中的质量安全控制问题,对分离自冷藏过程中发生腐败的黄河鲤鱼的一株优势腐败菌进行了鉴定和腐败特性研究。通过16S rRNA序列比对分析了该菌的种属,并测定了该菌的胞外蛋白酶活性、生物被膜和泳动性,采用生物传感器法测定了该菌的群体感应活性。结果表明:该菌为嗜水气单胞菌,命名为Aeromonas hydrophila YCR17(A.hydrophila YCR17);该菌能够产生胞外蛋白酶,并随培养时间的增加而增强;形成的生物被膜在48 h时达到最大,为高强度生物被膜,随后开始逐渐消解、脱落;可以在泳动性培养基表面进行运动扩散,说明A.hydrophila YCR17具有泳动性;诱导紫色杆菌CV026产生紫色素,具有AHLs介导的群体感应调控系统。研究结果为黄河鲤鱼的腐败机制及质量控制提供了理论基础。 To solve the quality and safety control of Yellow River carp during storage and transportation.The characteristics of a dominant spoilage bacterium isolated from spoiled Yellow River carp during refrigeration were studied.The bacterium was analyzed by 16S rRNA sequence alignment method.Spoilage characteristics of extracellular protease activity,biofilm formation and swimming motility of the strain were measured,as well as its quorum sensing activity by biosensor method.Results showed that the strain was identified as Aeromonas hydrophila YCR17(A.hydrophila YCR17),which can secrete extracellular protease,and increased with the increase of culture time.A.hydrophila YCR17 formed high-strength biofilms,reached to the maximum at 48 h,and then began to digest.A.hydrophila YCR17 spread around on the surface of agar plate,indicating its swimming ability.A.hydrophila YCR17 was detected to induce Chromobacterium violaceum 026 to produce purple pigment,which presented the existance of AHLs mediated quorum sensing(QS)regulation system.This result provides theoretical basis for studying the spoilage mechanism and quality control of Yellow River carp.
