温度胁迫对祁连山裸鲤幼鱼血清生化指标及代谢酶活性的影响 被引量:2

摘要 为了对祁连山裸鲤苗种培育、良种选育及养殖推广提供参考依据,试验采用5个温度梯度处理(8℃、13℃、18℃、23℃、28℃)的胁迫条件下测定幼鱼血清生化指标和代谢活性的方法,探索温度胁迫对祁连山裸鲤幼鱼血清生化指标及代谢酶活性的影响。本试验周期为5 d,分别在第1 d、3 d、5 d对祁连山裸鲤幼鱼进行尾静脉采血,对血清的总蛋白(TP)含量、葡萄糖(GLU)含量、甘油三脂(TG)含量、碱性磷酸酶(AKP)酶活性、过氧化氢酶(CAT)酶活性、谷丙转氨酶(GPT)酶活性、谷草转氨酶(GOT)酶活性和超氧化物歧化酶(SOD)酶活性等8项指标进行了测定。结果表明,祁连山裸鲤幼鱼血清生化指标中,18℃最适温度对照组在不同时间的TP、GLU、TG含量同8℃低温胁迫组及28℃高温胁迫组存在显著差异(P<0.05),且不同温度组的GLU、TG含量均随着试验的进行而下降。在血清代谢酶活性中,18℃最适温度对照组在不同时间的AKP、CAT、SOD活性显著高于8℃低温胁迫组及28℃高温胁迫组(P<0.05),而GPT、GOT活性则呈现低于其他试验组的结果。温度胁迫对祁连山裸鲤幼鱼的血清生化指标及代谢酶活性指标有显著的影响,其生长状况及代谢功能在18℃条件下最佳。在苗种培育及人工养殖生产中应控制好水温,避免温度剧变急性胁迫,减少幼鱼的应激反应。 In order to provide reference basis for Gymnocypris chilianensis juveniles seedling breeding and cultivation, this study performed to measure serum biochemical indicators and metabolic activity of young fish under five temperature stress condition(8℃,13℃,18℃ 23℃,28℃).This test cycle was 5 d, and Gymnocypris chilianensis juveniles tail vein blood of young fish was collected at 1 d, 3 d, 5 d respectively.The serum parameters of total protein(TP),glucose(GLU),triglyceride(TG),alkaline phosphatase(AKP),catalase(CAT),alanine aminotransferase(GPT),glutamic oxalacetic aminotransferase(GOT) and superoxide dismutase(SOD) were studied.The results showed as follows: Among the serum biochemical indexes of Gymnocypris chilianensis juveniles, the contents of TP,GLU and TG in 18℃ optimal temperature control group at different times were significantly different from those in 8℃ low temperature stress group and 28℃ high temperature stress group, and the contents of GLU and TG in different temperature groups decreased with the experiment time.Among the activities of serum metabolic enzymes, the activities of AKP,CAT and SOD in 18℃ optimal temperature control group were significantly higher than those in 8℃ low temperature stress group and 28℃ high temperature stress group at different times, while the activities of GPT and GOT were lower than those in other experimental groups.Temperature stress has significant effects on serum biochemical indexes and metabolic enzyme activity indexes of Gymnocypris chilianensis juveniles.The growth and metabolic function of the fish are best at 18℃.Water temperature should be controlled well in seedling cultivation and artificial breeding to avoid acute stress of temperature up heaval and reduce stress response of Gymnocypris chilianensis juveniles.
