金钟花总黄酮提取工艺及抗氧化活性研究 被引量:1

摘要 目的:优化金钟花中总黄酮提取工艺条件,并对金钟花中总黄酮清除DPPH自由基活性进行初步研究。方法:以金钟花总黄酮得率为考察指标,通过单因素和响应面实验,建立各因素与响应值之间的数学模型,确定最佳提取工艺;以芦丁和Vc做对照,通过对清除DPPH自由基能力测定,评价金钟花总黄酮抗氧化活性。结果:金钟花总黄酮最佳提取工艺:乙醇浓度90%,料液比1 g:88 mL,超声时间20 min,在此条件下金钟花中总黄酮的得率为101.17 mg·g^(-1);金钟花总黄酮IC_(50)=11.05 mg·L^(-1)、VcIC_(50)=3.26 mg·L^(-1)和芦丁IC_(50)=17.53 mg·L^(-1),清除DPPH自由基能力排序为Vc>金钟花总黄酮提取液>芦丁。结论:利用响应面法优化的金钟花总黄酮提取工艺得率高,金钟花具有较强的抗氧化活性,且其体外抗氧化能力在一定范围内随着总黄酮质量浓度的增加而增加。 Objective:To optimize the extraction process conditions of total flavonoids from Forsythia viridissima Lindl,and conduct a preliminary study on the scavenging activity of total flavonoids from Forsythia viridissima Lindl for scavenging DPPH free radicals.Methods:Taking the yield of total flavonoids from Forsythia viridissima Lindl as the investigation index,the mathematical model between each factor and the response value was established through single factor and response surface experiments,and the optimal extraction process was determined;Determination of free radical capacity,evaluation of the antioxidant activity of total flavonoids of Forsythia viridissima Lindl.Results:The optimal extraction process of total flavonoids from Forsythia viridissima Lindl:The concentration of ethanol is 90%,the ratio of solid to liquid is 1 g:88 mL,and the ultrasonic time is 20 min.Under these conditions,the yield of total flavonoids in Flos vulgaris is 101.17 mg·g^(-1);IC_(50)=11.05 mg·L^(-1),VcIC_(50)=3.26 mg·L^(-1) and rutin IC_(50)=17.53 mg·L^(-1),the scavenging ability of DPPH free radicals was ranked as Vc>Victoria total flavonoid extract>rutin.Conclusion:The extraction process of total flavonoids from Forsythia viridissima Lindl by response surface methodology has a high yield.Forsythia viridissima Lindl has strong antioxidant activity,and the in vitro antioxidant capacity increases with the increase of total flavonoids concentration within a certain range.
