摘要
目的探讨金丝桃苷对肺癌A549细胞增殖、侵袭、凋亡及丝裂原活化细胞外信号调节激酶(MEK)/细胞外信号调节激酶(ERK)信号通路的影响。方法将肺癌A549细胞分成对照组、顺铂组(1μg/mL)、金丝桃苷低浓度组(50μg/mL)、金丝桃苷高浓度组(100μg/mL);对照组正常培养肺癌A549细胞不进行任何干预,顺铂组、金丝桃苷低浓度组、金丝桃苷高浓度组分别给予培养肺癌A549细胞对应浓度的药物。培养48h后,细胞计数试剂盒-8、Transwell小室、流式细胞仪分别检测细胞增殖、侵袭、凋亡能力;荧光定量PCR、蛋白印迹法分别检测MEK、ERK信使核糖核酸(mRNA)和蛋白表达水平。结果与对照组比较,顺铂组、金丝桃苷低、高浓度组细胞增殖抑制率、细胞凋亡率升高,侵袭细胞数目、MEK、ERK mRNA和蛋白表达水平降低(P<0.05);与顺铂组比较,金丝桃苷低、高浓度组细胞增殖抑制率、细胞凋亡率降低,侵袭细胞数目、MEK、ERK mRNA和蛋白表达水平升高(P<0.05);与金丝桃苷低浓度组比较,金丝桃苷高浓度组细胞增殖抑制率、细胞凋亡率升高,侵袭细胞数目、MEK、ERK mRNA和蛋白表达水平降低(P<0.05)。结论金丝桃苷能明显抑制肺癌A549细胞增殖、侵袭,促进其凋亡;其机制可能与金丝桃苷抑制MEK/ERK信号通路的激活有关。
Objective To investigate the effects of Hyperoside on proliferation,invasion,apoptosis and mitogen activated extracellular signal regulated kinase(MEK)/extracellular signal regulated kinase(ERK)signaling pathway of lung cancer A549 cells.Methods Lung cancer A549 cells were divided into the control group,cisplatin group(1μg/mL),Hyperoside low concentration group(50μg/mL),and Hyperoside high concentration group(100μg/mL).The normal cultured lung cancer A549 cells in the control group were not intervened,while the Cisplatin group,Hyperoside low concentration group and Hyperoside high concentration group were given corresponding concentrations of drugs.After 48 hours of incubation,cell counting kit-8,trans-well chamber and flow cytometry were used to detect the ability of cell proliferation,invasion and apoptosis,respectively.The expression levels of MEK,ERK messenger RNA(mRNA)and protein were detected by fluorescence quantitative PCR and western blot,respectively.Results Compared with the control group,the cell proliferation inhibition rate and apoptosis rate increased in the cisplatin group,Hyperoside low and high concentration groups,while the number of invasive cells,the expression levels of MEK,ERK mRNA and protein decreased(P<0.05);Compared with the cisplatin group,the cell proliferation inhibition rate and apoptosis rate decreased in the Hyperoside low and high concentration groups,while the number of invasive cells,the expression levels of MEK,ERK mRNA and protein increased(P<0.05);Compared with the Hyperoside low concentration group,the cell proliferation inhibition rate and apoptosis rate increased in the Hyperoside high concentration group,while the number of invasive cells,the expression levels of MEK,ERK mRNA and protein decreased(P<0.05).Conclusion Hyperoside can significantly inhibit the proliferation and invasion of lung cancer A549 cells and promote apoptosis,and its underlying mechanism may be related to the inhibition of activation of MEK/ERK signaling pathway by Hyperoside.
