益脑通络颗粒对小鼠出凝血时间和大鼠凝血四项以及血细胞的影响

Effects of Yinao Tongluo Granule on Bleeding and Coagulation Time in Mice,Four Items of Coagulation and Blood Cells in Rats

摘要 目的:探讨益脑通络颗粒对小鼠出、凝血时间和大鼠凝血四项及血细胞的影响。方法:以ICR小鼠和SD大鼠为实验对象,随机分为空白对照组,益脑通络颗粒低、中、高剂量组,灌胃7 d后,采用毛细管法分别测定各组小鼠出、凝血时间;收集大鼠血液标本,检测凝血四项及血细胞指标,凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、纤维蛋白原(FIB)、红细胞(RBC)、血红蛋白(HGB)以及红细胞压积(HCT)。结果:与空白对照组相比,益脑通络颗粒低、中、高剂量组均能明显延长小鼠出、凝血时间(P<0.01);与空白对照组相比,益脑通络颗粒低剂量组的APTT显著延长(P<0.01),PT和TT差异无统计学意义(P>0.05),FIB指标明显下降(P<0.05);中剂量组APTT、PT均显著延长(P<0.01),TT明显延长(P<0.05),FIB指标显著下降(P<0.01);高剂量组PT显著延长(P<0.01),FIB指标明显下降(P<0.01),APTT和TT无显著变化(P>0.05);与空白对照组相比,益脑通络颗粒高剂量组可以降低大鼠红细胞、血红蛋白水平(P<0.05),并显著降低红细胞压积(P<0.01),低、中剂量组差异均无统计学意义(P>0.05)。结论:益脑通络颗粒具有良好抗凝血和改善血液循环的作用。 Objective:To investigate the effects of Yinao Tongluo Granule on bleeding and coagulation time in mice and four items of coagulation in rats.Methods:ICR mice and SD rats were randomly divided into the blank control group and the Yinao Tongluo Granule groups of low-dose,medium-dose and high-dose.After intragastric administration for 7days,the bleeding and coagulation time of mice were measured by capillary method in different groups.Blood samples of rats were collected to detect four items of coagulation and blood cells,including prothrombin time(PT),activated partial thromboplastin time(APTT),thrombin time(TT)and fibrinogen(FIB),red blood cells(RBC),hemoglobin(HGB)and hematocrit(HCT).Results:Compared with those in the blank control group,the bleeding and coagulation time of mice were significantly prolonged in the Yinao Tongluo Granule groups of low-dose,medium-dose and high-dose(P<0.01).Compared with those in the blank control group,APTT was significantly prolonged in the Yinao Tongluo Granule group of low-dose(P<0.01),there was no significant differences in PT and TT between the two groups(P>0.05),and FIB index decreased significantly in the Yinao Tongluo Granule group of low-dose(P<0.05);APTT and PT were significantly prolonged(P<0.01),TT was significantly prolonged(P<0.05),and FIB index was significantly decreased in the medium-dose group(P<0.01);PT was significantly prolonged(P<0.01),FIB index was significantly decreased(P<0.01),APTT and TT had no significant changes in the high-dose group(P>0.05).Compared with those in the blank control group,RBC and HGB were reduced(P<0.05),and HCT was significantly decreased in the high-dose group(P<0.01);of which there were no significant changes in the low-dose group and the medium-dose group(P>0.05).Conclusion:Yinao Tongluo Granule has good effects of anti-coagulation and improving blood circulation.
