正交优化柴胡地上部分总黄酮提取工艺及抑菌活性研究 被引量:1

Study on optimization of extraction technology and antimicrobial activity of total flavonoids from the aerial part of Bupleurum chinense by orthogonal design

摘要 目的正交优化柴胡地上部分总黄酮提取工艺,并测定其抑菌活性。方法以总黄酮提取率为指标,采用正交实验设计法,考察料液比、浸泡时间、提取时间、提取次数4个因素的提取最佳水平;以96孔板法对总黄酮进行4种细菌和4种真菌的抑菌活性测定。结果柴胡地上部分总黄酮的提取工艺最佳条件为:固液比1∶10,浸泡时间60 min,提取时间72 min,提取次数3次。通过验证在此条件下柴胡地上部分总黄酮提取率可达2.702%;抑菌活性的测定结果表明柴胡地上部分总黄酮对供试的4种细菌和4真菌均有一定的抑菌活性。结论该提取工艺简单可靠、合理可行,柴胡地上部分总黄酮具有一定的抑菌活性。 Objective To optimize the extraction process of total flavonoids from the aerial part of Bupleurum chinense by orthogonal experiment design and determining its bacteriostatic activity.Methods Based on one factor test,an orthogonal experimental design was employed,with total flavonoids extract ratio as an index,to optimize four extraction factors:solid-liquid ratio,soaking time,extraction time and times of extraction,the antimicrobial activities of four kinds of bacteria and four kinds of fungi were determined by 96-well plate method.Results The optimum technology conditions of extraction total flavonoids from the aerial part of Bupleurum chinense were as follows:ratio of solid to liquid of 1∶10,soaking time of 60 min,extraction time of 72 min,extraction times 3 times.The extraction rate of total flavonoids from the aerial part of Bupleurum chinense was 2.702%by these conditions.The results showed that the total flavonoids in the aerial part of Bupleurum chinense had certain antimicrobial activity against four bacteria and four fungi tested.Conclusion The extraction process is simple,reliable,reasonable and feasible.The total flavonoids in the aerial part of Bupleurum chinense had certain bacteriostatic activity.
