摘要
目的研究外周血中CD49a阳性NK细胞的功能及其在类风湿关节炎中的作用。方法流式细胞术检测健康志愿者和类风湿关节炎患者外周血NK细胞及其亚群包括CD56^(bright)NK、CD56^(dim)NK细胞群上CD49a及IFN-γ的表达;ELISA检测健康志愿者和类风湿关节炎患者血浆中IFN-γ的表达。结果与健康志愿者相比,RA患者外周血NK细胞及其2个亚群上CD49a的表达均显著升高。在RA患者血浆、外周血NK细胞及其亚群上IFN-γ的表达显著升高;同时CD49a与IFN-γ双阳性的NK细胞比例增加,且CD49a阳性NK细胞较CD49a阴性NK细胞表达更高水平的IFN-γ。结论CD49a阳性NK细胞参与类风湿关节炎疾病进程。
This study was designed to investigate the function of peripheral blood CD49a^(+) NK cells and their role in rheumatoid arthritis.Flow cytometry was used to detect the expression of CD49a and IFN-γin total NK cells and its subsets including CD56^(bright) NK and CD56^(dim) NK cells from peripheral blood of healthy controls and rheumatoid arthritis patients;ELISA was used to detect the expression of IFN-γin plasma of the healthy controls and rheumatoid arthritis patients.Compared with the healthy controls,the expression of CD49a in peripheral blood NK cells and its two subsets from rheumatoid arthritis patients were significantly increased,and IFN-γexpression also increased significantly in rheumatoid arthritis patients’plasma,total NK cells and its two subsets.Furthermore,CD49a and IFN-γdouble positive NK cells were increased,and CD49a positive NK cells expressed a higher level of IFN-γthan CD49a negative NK cells.In conclusion,CD49a positive NK cells are involved in the rheumatoid arthritis disease process.
