中泰南五味子转录组测序及生物信息学分析 被引量:1

摘要 【目的】利用高通量测序技术获得中泰南五味子的转录组信息。【方法】中泰南五味子叶片总RNA经过提取、建库、测序、de novo组装,获得Unigene序列,再进行注释和比对,得到中泰南五味子转录组的遗传信息。【结果】共获得29915条Unigene,平均长度为1033 bp,N50为1554 bp。共有22359条Unigene在至少1个数据库中获得功能注释,中泰南五味子与荷花Nelumbo nucifera的序列相似度最高。在京都基因与基因组百科全书(KEGG)数据库中共有14284个Unigene得到注释;在基因本体论(GO)数据库中共有18084条中泰南五味子Unigene获得注释,包括ATP结合、膜的组成成分和生物过程等。此外,采用MISA对组装的1000 bp以上的10747条Unigene进行简单序列重复(SSR)检测,共鉴定到6363个SSR位点,并使用Primer 3.0设计引物。【结论】获得的转录组数据为中泰南五味子的药用活性成分合成通路和功能基因的挖掘奠定了基础,也有利于其资源保护和开发利用。 Objective To obtain transcriptomic information of Kadsura ananosma Kerr by high-throughput sequencing technology.Methods The total RNA of Kadsura ananosma Kerr leaves was extracted,library built,sequenced and de novo assembled to obtain unigene sequences,which were then annotated and compared to obtain the genetic information of the Kadsura ananosma Kerr transcriptome.Results A total of 29915 unigenes were obtained with an average length of 1033 bp and an N50 of 1554 bp.A total of 22359 unigenes were functionally annotated in at least one database,with the highest sequence similarity between Kadsura ananosma Kerr and Nelumbo nucifera.A total of 14284 unigenes were annotated in the Kyoto Encyclopedia of Genes and Genomes(KEGG)database;and a total of 18084 Kadsura ananosma Kerr unigenes were annotated in the Gene Ontology(GO)database,including package ATP binding,membrane constituents and biological processes.In addition,10747 unigenes assembled above 1000 bp were subjected to simple sequence repeat(SSR)detection using MISA,and a total of 6363 SSR sites were identified and primers were designed using Primer 3.0.Conclusion The achieved transcriptome datas provide a basis for the excavation of the synthetic pathways and functional genes of the medicinal active ingredients of Kadsura ananosma kerr,and also facilitate the conservation and exploitation of its resources.
