摘要
目的 应用高效液相色谱-电喷雾质谱(HPLC-ESI-MS^(n))联用技术对流苏石斛中主要的黄酮成分进行分析鉴定。方法 采用Kromasil 100-5 C色谱柱,用乙腈-0.1%甲酸溶液为流动相进行梯度洗脱以分离各主要成分,体积流量为0.8 mL·min,柱温为35℃,紫外检测波长为340 nm;质谱使用ESI离子源,负离子模式下采集数据。结果 结合相关文献及碎片离子信息进行数据分析,从流苏石斛中共鉴定出黄酮类化学成分27个,均为黄酮碳苷。该研究发现流苏石斛碳苷取代糖为葡萄糖、阿拉伯糖与1个未知己糖为主,3个芹菜素-6,8-二-C-己糖苷(峰1、峰2、峰4)相对丰度均较大,具有鉴别意义。结论 该研究可为流苏石斛的质量控制提供参考。
Objective High-performance liquid chromatography coupled with electrospray ionization multi-stage tandem mass spectrometry(HPLC/ESI-MS^(n))was used to identify the flavonoids in Dendrobium fimbriatum Hook..Methods HPLC analysis was performed with a Kromasil 100-5 Ccolumn. Gradient elution was performed with a mobile phase consisting of acetonitrile and 0.1% formic acid in water at a flow rate of 0.8 mL·min. The column temperature was maintained at 35 ℃. The detection wavelength was 340 nm. ESI ion source was used for mass spectrometry, and data were collected in negative ion mode. Results A total of 27 C-glycosyl flavones were identified in Dendrobium fimbriatum Hook.. The results were obtained by combining relevant literatures and fragment ion information with data analysis. In this study, it was found that glucose, arabinose and one unfriend sugar were the main substituents of carboside in Dendrobium fimbriatum Hook.. The relative abundance of three apigenin 6,8-di-c glycosides(peak1,peak2,peak4) was relatively high which was of significance for identification. Conclusion This study provided reference for quality control of Dendrobium fimbriatum Hook..
